摘要
目的:研究热量控制对肥胖大鼠白色脂肪组织(WAT)中瘦素以及糖脂代谢的影响。方法:Wistar雄性大鼠10只为正常组(N组)给予普通饲料喂养,另20只高脂饲料喂养造模,造模成功后通过随机数字表法分为模型组(M组)和热量控制组(CR组),各10只。除CR组每只给予M组同龄大鼠摄食量的70%,其他两组大鼠均自由进食饮水,共干预8周。检测各组大鼠体质量和血清葡萄糖(FPG)、胰岛素(INS)、总胆固醇(TC)、甘油三酯(TG)及瘦素的含量,并检测WAT中瘦素的蛋白表达。结果:干预8周后,与M组相比,CR组体质量、血清FPG、INS、TG、TC及瘦素含量均显著下降(P<0.01),且WAT中瘦素蛋白的表达水平显著降低(P<0.01)。结论:热量控制能减轻肥胖大鼠体质量,改善糖脂代谢,降低血清瘦素水平,下调WAT中瘦素的蛋白表达。
Objective:To observe the effect of caloric restriction on the expression of leptin in white adipose tissue(WAT)and glycolipid metabolism of obese rats.Methods:A total of 10 male Wistar rats as normal control group(N group)fed with normal diet,the other 20 rats with obesity induced with a high-fat diet were randomly divided into model group(M group)and caloric restriction group(CR group),each group were 10 rats.The CR group was fed only 70%of high-fat diet in the M group,the rats of N group and M group were fed with free eating and drinking,the intervention lasted for 8 weeks.The rats’body weight and the level of serum FPG,INS,TG,TC,leptin,and the expression of leptin in WAT were detected.Results:Compared with M group,body weight and the level of serum FPG,INS,TG,TC and leptin were significantly decreased in CR group(P<0.01).And the expression of leptin in the WAT decreased significantly in CR group(P<0.01).Conclusion:Caloric restriction can effectively reduce the body weight and regulate glycolipid metabolism of obese rats.Besides,it also can reduce the level of serum leptin and the expression of leptin in WAT.
