摘要
目的:探讨标本配穴预处理对糖尿病大鼠心肌缺血再灌注损伤炎症因子的调控机制。方法:选用3月龄Wistar大鼠50只,用随机数字法将50只大鼠随机分为糖尿病假手术组(DS组)、糖尿病缺血再灌注组(DMI组)、糖尿病电针预处理组(DEA组)、糖尿病电针+抑制剂预处理组(DEA+DSB组)、糖尿病抑制剂预处理组(DSB组)。采用腹腔注射链脲佐菌素建立糖尿病大鼠模型,成功后行左冠状动脉推管造模术,建立心肌缺血再灌注损伤模型,检测心肌超氧化物歧化酶(SOD)活性,心肌组织丙二醛(MDA)、IL-6、IL-1β、IL-10含量,透射电镜下观察心肌超微组织的变化。结果:DMI组、DEA组和DEA+DSB组、DSB组的IL-6、IL-1β和IL-10较DS组均显著升高(P<0.05)。DEA组、DEA+DSB和DSB组的IL-6和IL-1β较DMI组均显著降低(P<0.05),IL-10较DMI组均升高(P<0.05)。DS组、DEA组、DEA+DSB组和DSB组的大鼠心肌SOD含量较DMI组显著上升(P<0.05),MDA含量降低(P<0.05)。与DS组比较,DMI组心肌肌纤维溶解断裂,且DEA组及DEA+DSB组心肌肌纤维间隙增宽较小,心肌细胞肿胀减轻且坏死灶改善。结论:标本配穴预处理能调节糖尿病大鼠心肌缺血再灌注损伤模型的相关炎症细胞因子的表达,减轻氧化应激反应。
Objective:To investigate the regulation mechanism of points combination of manifestation and root cause pretreatment on myocardial ischemia-reperfusion injury inflammatory factors in diabetic rats.Methods:A total of 50 Wistar rats aged 3 months,were randomly divided into diabetic sham operation group(DS)and diabetic ischemia-reperfusion group(DMI)by random number method.Diabetes electroacupuncture pretreatment group(DEA),diabetic electroacupuncture+inhibitor pretreatment group(DEA+DSB),diabetes inhibitor pretreatment group(DSB).A diabetic rat model was established by intraperitoneal injection of streptozotocin.After successful left coronary artery catheterization,a model of myocardial ischemiareperfusion injury was established to detect myocardial superoxide dismutase(SOD)activity.Aldehyde(MDA),myocardial tissue IL-6,IL-1β,IL-10,and other indicators.Transmission electron microscopy was used to observe the myocardial ultrastructural changes.Results:IL-6,IL-1β,and IL-10 in the DMI group,DEA group and DEA+DSB group and DSB group were significantly higher than those in the DS group(P<0.05).The levels of IL-6 and IL-1βin the DEA group,DEA+DSB,and DSB groups were significantly lower than those in the DMI group(P<0.05).IL-10 in the DEA group,DEA+SB group,and DSB group were higher than those in the DMI group(P<0.05),and the difference was statistically significant.The myocardial SOD content in the DS group,DEA group,DEA+DSB group,and DSB group was significantly higher than that in the DMI group(P<0.05),and the MDA content was decreased(P<0.05).The difference was statistically significant.Compared with DS group,myocardial fibers in DMI group dissolved and fractured,and myocardial fiber gap widened less in DEA group and DEA+DSB group,myocardial cell swelling decreased and necrotic foci improved.Conclusion:Pretreatment with points combination of manifestation and the root cause can regulate the expression of related inflammatory cytokines and reduce oxidative stress in myocardial ischemiareperfusion injury model in diabetic rats.
