摘要
采用索氏提取法提取苹果皮总黄酮,研究了苹果皮总黄酮的最佳提取工艺及含量测定方法。正交实验确定了苹果皮总黄酮在300 W超声波辅助下的最佳提取工艺条件为:乙醇体积分数60%、料液比1:20(g:mL)、提取时间50min、提取温度50℃。以AlCl_3为显色剂、芦丁为标准品,采用紫外可见分光光度法在423nm波长处测定所生成络合物的吸光度,绘制芦丁标准曲线,拟合线性回归方程为:A=27.78c+0.0004,相关系数R^2=0.9980。按此方法分析苹果皮样品,测得苹果皮总黄酮含量为26.94mg·g^(-1)(n=6,RSD=0.92%)。该方法简单易操作,可为水果蔬菜中总黄酮含量的测定提供理论基础。
Total flavonoids from apple peels were extracted with a Soxhlet extraction method.The extraction conditions were optimized by orthogonal experiment,and determination method of content of total flavonoids was studied.The optimal extraction conditions were as follows:60%ethanol,solid-liquid ratio 1:20(g∶mL),treating with ultrasonic power of 300 Wfor 50min,and extraction temperature of 50℃.Using AlCl_3 as a chormogenic reagent,rutin as standard,absorbance of the complex was measured at 423 nm by UV-Vis spectrophotometry.Calibration curve of rutin was drawed,and linear regression equation of A=27.78c+0.0004(R^2=0.9980)was obtained.Applying the proposed method,the content of total flavonoids from apple peels was 26.94mg·g^(-1)(n=6,RSD=0.92%).The method is easy to operate,which provides a theoretical basis for the determination of total flavonoids from fruits and vegetables.
