摘要
目的:探讨阿托伐他汀预处理对垂体后叶素(Pituitrin,pit)致大鼠冠脉痉挛的作用。方法:40只SD大鼠随机分为四组,即对照组(不处理),阿托伐他汀组[5 mg/(kg·d)],硝苯地平组[5 mg/(kg·d)]和联合用药组(阿托伐他汀组+硝苯地平组),灌胃15 d后,经鼠尾静脉注入Pit 1.5 U/kg,诱发冠脉痉挛。观察注射Pit前(0 s)、注射后30 s、1、3、5和7 min时间点标Ⅱ导联J点变化、心律失常的发生率及心率。结果:注射Pit后30 s阿托伐他汀组不能抑制J点压低,硝苯地平组和联合用药组能抑制J点压低(P<0.05);注射Pit后1 min时阿托伐他汀组和硝苯地平组不能抑制J点压低,联合用药组能抑制J电压低(P<0.05);3、5、7 min三组均能抑制J点压低(P>0.05)。Pit诱发冠脉痉挛后出现的心律失常,阿托伐他汀组心律失常发生率与对照组一样(都为60%),硝苯地平较对照组低(20%),而联合组未发生心律失常(0)。各组间心率无明显差异。结论:单独应用阿托伐他汀能抑制J点压低,但阿托伐他汀联合硝苯地平能更好地预防大鼠冠脉痉挛的发生,显著降低心律失常的发生率。
Objective:To explore the effect of pretreatment with atorvastatin on Pituitrin(Pit)-induced coronary artery spasm of rats.Method:Forty male SD rats were randomly divided into four groups.Every group had 10 rats.The control group was non-drug intervention.The Atorvastatin group was given orally Atorvastatin[5 mg/(kg·d)] by gastric gavage for 15 days.The Nifedipine group was given orally Nifedipine[5 mg/(kg·d)] by gastric gavage for 15 days.The combined group was given orally Atorvastatin[5 mg/(kg·d)] and Nifedipine[5 mg/(kg·d)] by gastric gavage for 15 days.After 15 days continuous administration to all the groups of rats,we created the coronary artery spasm by injecting Pit into caudal vein of rats.To monitor the Ⅱ Load Electrocardiogram and record the Cardiographs of all the groups at the time of pre-injection(0 s),post-injection(30 s,l,3,5,7 min) of the Pit.Compared the change range of the J point,the arrhythmogenesis rate and heart rate at all of the timing points.Result:After injecting Pit 30 s,the Atorvastatin group didn’t reduce the depress of J point,but the Nifedipine group and the combined group resisted J-point changes(P〈0.05).After injecting pit 1 min,the Atorvastatin group and the Nifedipine group didn’t reduce the depress of J point,but the combined group decreased J-point changes(P〈0.05).After injecting Pit 3,5,7 min,all reduced the variation(P〉0.05).In comparing of the arrhythmogenesis rate of all groups,there were 6 rats(60%) in the control group,in the Atorvastatin group,the Nifedipine group and the combined group the arrhythmogenesis rate were accordingly 6(60%),2(20%) and 0.There’s no obvious difference between the administrated groups and the model group for the heart rate(P〉0.05).Conclusion:Atorvastatin can evidently improve J-point changes of ECG of rats induced by Pit,but the addition of Atorvastatin to Nifedipine can better prevent coronary spasm,and decrease the arrhythmogenesis rate.
