摘要 目的 观察咖啡因对体外培养U251神经胶质瘤细胞存活及凋亡的影响。方法 采用MTT和AnnexinV/PI流式细胞法观察不同浓度和作用时间的咖啡因对U251神经胶质瘤细胞的影响。结果 (1)咖啡因在体外可以抑制U251人胶质瘤细胞的存活;(2)咖啡因在体外使U251细胞早期凋亡发生的时间较晚,且在1~10mmol/L浓度范围内,出现浓度依赖性瘤细胞早期凋亡增加,大于10mmol/L时,瘤细胞迅速出现大量坏死。结论 实验初步证明了咖啡因在体外可以抑制U251人胶质瘤细胞的增殖活性并在一定浓度范围内(1~10mmol/L)可促使其凋亡,为进一步的动物实验及新药开发提供实验依据。 Purpose To investigate the effects of caffeine on cell proliferation and apoptosis of human glioma cell line U251 in vitro. Methods MTT and Annexin V/PI flow cytometry were employed undergoing different caffeine concentrations and time on U251 glioma cells. Results (1) The cell viability of U251 was significantly surpressed by caffeine in vitro. (2) In vitro, the apoptosis time of U251 cell is late under caffeine, and caffeine concentration during 1 - 10 mmol/L is effective, however, exceeding 10 mmol/L, showed a lot of necrotic U251 cells rapidly. Conclusions The experiment proved that the caffeine-treated U251 cell is suppressed proliferation and caffeine can induce the apoptosis of treated U251 cell in a certain range of concentration ( 1 - 10 mmol/L) , so as to provide experimental basis for further animal experiments and the development of new drugs.
