摘要
目的观察穴位埋线"足三里"对单纯性肥胖小鼠的减肥效应和脂肪组织中IL-6、TNF-α与MCP-1基因表达变化的影响。方法选用30只刚断乳的健康清洁级C57BL/6J雄性小鼠,随机挑选6只普通饲料喂养组为正常组,另一组24只高脂饲料喂养,将造模成功的12只随机分为模型组和埋线组。比较各组小鼠体重、体脂、血脂、血糖等相关指标的差异,用HE染色观察比较各组小鼠脂肪的形态学差异,用实时荧光定量PCR检测小鼠脂肪IL-6、TNF-α与MCP-1表达变化。结果治疗后,埋线组小鼠体重下降明显,内脏脂肪量与模型组相比,差异明显。血脂方面,埋线组与模型组相比,胆固醇、低密度脂蛋白、甘油三酯和葡萄糖含量含量均显著降低。埋线组小鼠脂肪组织IL-6、TNF-α、MCP-1的mRNA表达水平较模型组显著降低。结论穴位埋线"足三里"可以降低肥胖小鼠的体重,降低小鼠内脏脂肪含量和血脂、血糖水平,抑制肥胖小鼠脂肪组织炎症因子IL-6-mRNA、TNF-α-mRNA与MCP-1-mRNA的表达,减轻炎症反应,从而达到减脂的目的。
Objective To observe the effects of acupoint catgut- embedding therapy on fat reducing and expressions of IL- 6,TNF- α,MCP- 1 in adipose tissue of the obese mice. Methods Choose 30 healthy and clean weaned C57 BL / 6J male mice,and then randomly divide them into normal group and high- fat group,the normal group with normal diet in 6,high- fat group with high fat diet in 24. After successful modeling,the 12 obese mice are randomly divided into model group and acupoint catgut- embedding therapy( ACE) group. Compare the differences in mice weight,fat,blood sugar,blood fat of each group,the fat morphological differences among groups with HE staining,using real- time PCR to detect the expression of adipose IL- 6,TNF- α,MCP- 1. Results After treatment,the ACE group of mice significantly decreased body weight,the visceral fat mass compared with the model group. The aspect of blood fat,the level of cholesterol,low- density lipoprotein,triglycerides and glucose,the ACE group decreased significantly compared with the model group. The ACE group’s the expression of adipose tissue IL- 6-mRNA,TNF- α- mRNA,MCP- 1- mRNA are significantly decreased compared with the model group. Conclusion Acupoint catgut- embedding " Zusanli" can reduce weight,visceral fat mass,blood fat,blood sugar levels of the obese mice and inhibit the inflammatory factor in adipose tissue of obese mice,such as IL- 6,TNF- α,MCP- 1. The effects of acupoint catgut- embedding therapy on fat reduction might be related to the inflammatory factor.
