基于细胞色素C氧化酶基因(COI)和核糖体16S rDNA条形码的中药海蛇品种鉴定 被引量:5

摘要 海蛇是我国名贵药材之一,新鲜捕捞的海蛇不易保存,一般立刻晒制保存,晒干后海蛇外观变化大,不宜再用外观鉴定,而基因条形码作为一种较新的分子鉴定方法则不再受外观变化困扰,适用于海蛇晒干制品的品种鉴定。该以平颌海蛇Lapemis hardwickii、环纹海蛇Hydrophis fasciatus、尖尾海蛇Aipysurus eydouxii、贝尔切海蛇H.belcheri和兰伯特海蛇H.lamberti等5种海蛇为研究对象,通过提取实验和基因测序技术测定了这5种海蛇的COI和16S r DNA基因序列,对这些序列进行互相比对并用相似性搜索算法评价5条序列的鉴定成功率。研究显示,16S r DNA基因条形码能初步区分海蛇种属,COI基因条形码序列种内种间差异明显,能进一步区分海蛇品种。因此表明COI序列可以作为海蛇晒干制品的品种鉴定方法。 Hydrophidae,one of the precious traditional Chinese medicines,is generally drily preserved to prevent corruption,but it is hard to identify the species of Hydrophidae through the appearance because of the change due to the drying process. The identification through analysis on gene barcode,a new technique in species identification,can avoid this problem. The gene barcodes of the 5species of Hydrophidae,Lapemis hardwickii,Hydrophis fasciatus,Aipysurus eydouxii,Hydrophis belcher and Hydrophis lamberti,were acquired through DNA extraction and gene sequencing. These barcodes were then in sequence alignment and test the identification efficiency by BLAST. Our results showed that the 16 S r DNA sequences identified Hydrophidae briefly and the COI sequenceshad obvious difference between intra-and inter-species,indicating that DNA bar-coding was an efficiency method of Hydrophidae identification.
