摘要
目的阐明瞬时受体电位离子通道家族(TRP)中香草素受体亚家族(TRPV)成员TRPV4和小电导钙激活钾通道3(SK3)复合物对老年大鼠气管平滑肌收缩的影响。方法运用离体气管张力实验,观察高钾、卡巴胆碱对年轻、老年大鼠气管收缩的影响,以及瞬时受体电位离子通道TRPV4特异性激动剂和抑制剂对卡巴胆碱引起的气管收缩的影响;运用Western blot法检测TRPV4和SK3在年轻、老年大鼠气管平滑肌上表达量的变化。结果与年轻大鼠比较,老年大鼠高钾引起的气管收缩没有显著改变,但卡巴胆碱引起的收缩显著增强,且TRPV4激动剂GSK引起的舒张反应显著降低;两组大鼠用TRPV4阻断剂HC或RN1734预处理后,GSK引起的舒张反应均被显著阻断;Western blot结果显示,与年轻大鼠比较,老年大鼠气管平滑肌组织TRPV4表达显著升高,SK3表达却显著降低。结论 TRPV4-SK3信号复合物在老年大鼠气管平滑肌表达改变可能与老年大鼠气管舒张效应减弱有密切关系。
Objective To investigate the functional role of transient receptor potential vanilloid 4( TRPV4)-small conductance Ca2 +activated potassium channel 3( SK3) in the contraction of aged rat airway smooth muscle. Methods We used tracheal tension measurement to observe the effect of high K+solution and carbachol on the tracheal contraction and the effect of TRPV4 channel agonist( GSK) and antagonist( HC and RN1734) on carbachol-induced tracheal contraction. Western blot was used to examine the expression level of TRPV4 and SK3. Results Compared with the young rats,high K+solution-induced contraction did not change in the aged rat tracheal rings. However,carbachol-induced contraction was significantly increased. The relaxation induced by GSK was notably reduced in the aged rat tracheal rings. On the other hand,TRPV4 antagonist HC and RN1734 significantly inhibited GSK-induced relaxation. Western blot data showed that the expression level of TRPV4 was markedly increased; but SK3 expression was significantly decreased in the aged rat airway smooth muscle. Conclusion The change of the expression level of TRPV4-SK3 complex may be the reason for the reduction of tracheal relaxation.
