宫颈癌组织HeLa细胞RNF43表达及其对宫颈癌细胞增殖能力的影响 被引量:3

摘要 目的观察RNF43在宫颈癌组织中的表达水平及其与宫颈癌发生发展的相关性。方法纳入2013年1月—2015年1月华北石油管理局总医院妇产科诊治的76对配对的宫颈癌及癌旁组织标本为研究对象,提取组织总RNA,应用半定量RT-PCR方法检测RNF43的表达水平,结合宫颈癌患者临床资料分析RNF43表达与宫颈癌患者临床资料之间的相关性。在5株宫颈癌细胞株中检测RNF43的表达,并通过MTT增殖实验和克隆形成实验检测RNF43对宫颈癌细胞株增殖的影响。结果半定量RT-PCR发现60.5%(46/76)的宫颈癌中RNF43低表达,且RNF43在宫颈癌中的低表达与肿瘤的TNM分期以及淋巴结转移相关(X^2=15.134、13.740,P<0.01)。RNF43在HeLa细胞中无表达,在SiHa细胞和CaSki细胞中低表达,在C-33A和C4-1细胞中高表达。在HeLa细胞中恢复RNF43表达后可以抑制细胞的增殖和克隆形成能力。结论 RNF43在宫颈癌中频发表达降低,且与肿瘤的TNM分期及淋巴结转移相关,RNF43具有作为宫颈癌诊断及预后评估标志物的潜能,可抑制宫颈癌细胞的增殖能力。 Objective To observe the expression level of RNF43 in cervical cancer tissue and its correlation with the development of cervical cancer.Methods From January 2013 to January 2015,76 cases of matched cervical cancer and adjacent tissue were enrolled in this study.Total RNA of tissues was extracted,and semiquantitative RT-PCR was used in this study to detecte the expression level of RNF43,then analysed the correlation between the expression level of RNF43 and cervical cancer patients' clinical materials.The expression of RNF43 was detected in 5 cervical cancer cell lines by RT-PCR and the effect of RNF43 on cervical cancer cells was detected by MTT proliferation assay and colony formation.Results RT-PCR proved that RNF43 expressed at low level in 60.5%(46/76) cervical cancer.And the low level of RNF43 expression was significantly associated with TNM stage and lymph node metastasis(χ-2=15.134,13.740,P〈0.01).No expression of RNF43 was found in HeLa cells,low level expression was found in SiHa and CaSki cells,and highly expressed in C-33 A and C4-1 cells.Restoration of RNF43 expression in HeLa cells can inhibit the proliferation and colony formation abilities.Conclusion The RNF43 protein is frequently lower expressed in cervical cancer and associated with TNM stage and lymph node metastasis.RNF43 may serve as a diagnosis and prognostic biomarker in cervical cancer and can inhibit the proliferation of cervical cancer cells.
