甘肃不同产地黄芩提取物的抗肿瘤作用研究 被引量:2

摘要 目的:研究甘肃不同产地黄芩提取物对肝癌H_(22)模型小鼠的抑瘤作用。方法:将H_(22)瘤株接种于小鼠,建立移植瘤模型。将实验动物分为正常组,模型组,5-Fu组,陇西、漳县、渭源、岷县产黄芩提取物组。于小鼠接种H_(22)瘤株第2 d开始给药,黄芩组每d每只灌服黄芩提取物6 g/kg,每d1次,5-Fu组按25 mg/kg剂量隔日给药1次,正常对照组和模型组每d每只给予0.2 m L/10 g的生理盐水,给药10 d。观察给药后小鼠体质量变化,计算肿瘤抑制率;ELISA法检测血清白细胞介素-2(IL-2)和肿瘤坏死因子-α(TNF-α)含量;比色分析法检测血清T-AOC活力和MDA含量;单细胞凝胶电泳检测外周血淋巴细胞DNA损伤程度。结果:不同产地黄芩提取物对肿瘤的生长均有一定的抑制作用,抑瘤效果:陇西产黄芩提取物>漳县产黄芩提取物>渭源产黄芩提取物>岷县产黄芩提取物;同时,明显升高移植瘤小鼠血清IL-2水平和降低TNF-α水平,升高血清T-AOC活力和降低MDA含量,减轻外周血淋巴细胞DNA损伤。结论:不同产地的黄芩提取物不但可以抑制肿瘤的生长,而且可以通过调节IL-2和TNF-α等细胞因子的表达,改善或增强机体的免疫功能,以及通过提高机体的抗氧化能力,减轻自由基对脂质、淋巴细胞DNA的过氧化损伤,发挥其抗肿瘤作用。 Objective: To study the inhibition effect of Scutellaria baicalensis Georgi extracts from different areas of Gansu Province on H22 transplanted tumor in mice. Methods: The H22 transplanted mouse models were established. Mice were randomly divided into 7 groups: normal,model,5- Fu positive control,Scutellaria baicalensis Georgi extracts from Longxi,Zhangxian,Minxian and Weiyuan groups. Two days after being inoculated with H22 tumor cells,mice were treated with 6 g / kg extracts from Scutellaria baicalensis Georgi once daily for 10 d,and with 25 mg / kg 5- Fu every other day for five times. The model and normal control groups were administrated with 0. 2 m L /10 g saline,then the body weight change and tumor growth inhibitory rate were calculated; the serum interlekin- 2( IL- 2) and tumor necrosis factor- α( TNF- α) levels were determined with ELISA method; the activity of T- AOC and the content of MDA in serum were detected by colorimetric method,as well as DNA damage of peripheral blood lymphocytes was observed by single cell gel electrophoresis( SCGE).Results: Extracts of Scutellaria baicalensis Georgi from different areas of Gansu Province inhibited the growth of transplanted tumor at different degree( Longxi 〉Zhang county 〉Weiyuan〉 Min county),in addition,extracts could elevated the serum IL- 2 level,reduced the serum TNF- α level,promoted the T- AOC activity and decreased the MDA content in serum,as well as notably reduced DNA damage of peripheral blood lymphocytes. Conclusion: Extracts from Scutellaria baicalensis Georgi have an anti- tumor effect on H22 transplanted tumor in mice,regulating the IL- 2 and TNF- α expressions to raise immunologic function; as well as improving anti- oxidant capability of H22- bearing mice to light peroxidative damages of lipid and DNA from free radicals,all maybe the mechanisms of extracts from Scutellaria baicalensis Georgi. on H22 cells.
