摘要
目的观察柔肝抑纤饮对实验性肝纤维化大鼠肝脏TGF-β_1、Smad4表达的影响。方法 60只Wistar大鼠除空白对照组外,余建立CCL4性肝纤维化模型,随机分为模型组、柔肝抑纤饮组和复方鳖甲软肝片组。实验8周后,处死动物,检测血清ALT、AST及肝组织匀浆HA、PCⅢ水平。行肝组织Masson染色及TGFβ_1、Smad4免疫组化染色,阳性结果计算机定量分析。结果模型组大鼠血清ALT、AST,肝匀浆HA、PCⅢ,以及肝组织Masson染色阳性面密度和Smad4、TGFβ_1免疫组化阳性率均较空白对照组明显升高(P<0.05,P<0.01);而柔肝抑纤饮组大鼠除血清ALT外,余指标均较模型组降低(P<0.05,P<0.01)。结论柔肝抑纤饮有明显的抗肝纤维化作用,其作用机制之一可能在于抑制TGF-β_1、Smad4的表达。
Objective To observe the effect of Rougan Yixian decoction on the expression of TGF-beta1 and Smad4 in experimental rat hepatic fibrosis. Methods The liver fibrosis rat model was induced with carbon tertrachloride and besides model group. The rats of liver firbosis were divided into model group, Rougan Yixian decoction group andcompond Biejia Ruangan tablet group. After 8weeks, all rats were killed, and the indexes were tested in blood and liver. The positive results were quantitative analysis. Results Comparing with the control group, the indexes in the model group of the content of ALT, AST in serum, the level of HA and PC Ⅲ in liver, the positive areal density of liver college and immunohistochemical positive rate of Smad4 and TGF β1were all higher(P0.05,P0.01). In addition to the serum ALT, the others in Rougan Yixian decoction group were lower than those in the model group( P0.05, P〈0.01). Conclusion Rougan Yixian decoction had obviously curative effect on anti-liver fibrosis. The one of mechanism of action was to control expression of the TGF-β1and Smad4.
