摘要
目的:观察健肾助孕方对着床障碍小鼠血清雌孕激素浓度和子宫HOXA10表达的影响。方法:将30只孕鼠随机分为模型组3只、健肾助孕方低剂量组6只、健肾助孕方高剂量组6只、黄体酮组7只和正常对照组8只。从妊娠第1天开始,模型组、正常对照组每日给予生理盐水0.2 m L·只-1灌胃,黄体酮组皮下注射黄体酮溶液0.1 m L·只-1,健肾助孕方低剂量组、高剂量组每日灌胃0.2 m L·只-1。妊娠第4天皮下注射米非司酮诱导着床障碍小鼠模型。妊娠第8天处死小鼠,ELISA法检测血清中雌孕激素浓度,免疫组织化学法和Western blot法对子宫中HOXA10的表达和蛋白含量进行检测。结果:健肾助孕方低剂量组、高剂量组血清雌孕激素浓度较模型组明显升高,差异有统计学意义(P<0.05)。健肾助孕方高剂量组、黄体酮组子宫中HOXA10的表达显著增多,与模型组比较,差异有统计学意义(P<0.05);健肾助孕方低剂量组、高剂量组、黄体酮组子宫中HOXA10的蛋白含量明显高于模型组,差异有统计学意义(P<0.05)。结论:健肾助孕方可以升调着床障碍小鼠血清雌孕激素水平,提高子宫中HOXA10的表达量,从而改善着床障碍小鼠的子宫内膜容受性,促进胚胎着床。
Objective: To observe the influence of Cai’ s Jianshen Zhuyun Decoction( CJSZYD) on estrogen and progesterone and womb HOXA10 expression of mice with imbed barrier. Methods: Thirty pregnant mice were randomly divided into the model group with 3 cases,the group of low-dosage CJSZYD with 6 cases,the group of high-dosage CJSZYD with 6 cases,the progesterone group with 7 cases and the normal control group with 8 cases. From the 1st day of pregnancy,any one in the model group and the normal control group were given 0. 2 m L of normal saline for gavage each day; Any one in the progesterone group were given 0. 1 m L of progesterone fluid for subcutaneous injection each day; Any one the group of low-dosage CJSZYD and the group of high-dosage CJSZYD were given 0. 2 m L of decoction for gavage. From the 4th day of pregnancy,mifepristone was used for subcutaneous injection in order to induce the mouse’s model with imbed barrier. On the 8th day of pregnancy,the mice were killed. Then serum estrogen and progesterone concentration was detected by using ELISA method; Womb HOXA10 expression and protein content were measured by using immunohistochemical method and Western blot method. Results: Compared with the model group,serum estrogen and progesterone concentration of the group of low-dosage CJSZYD and the group of high-dosage CJSZYD were obviously increased,and the differences between them had statistical significance( P 〈 0. 05). Compared with the model group,Womb HOXA10 expression the group of high-dosage CJSZYD and the progesterone group obviously increased,and the differences between them had statistical significance( P 〈 0. 05). Womb HOXA10 protein contents of the group of low-dosage CJSZYD and the group of high-dosage CJSZYD and the progesterone group were obviously higher than those of the model group,and the differences between them had statistical significance( P 〈 0. 05). Conclusion: CJSZYD can increase and regulate estrogen and progesterone levels and womb HOXA10 expression of mice with imbed barrier,thus improving endometrial receptivity and embryo implantation.
