6株破囊壶菌的分离鉴定及脂肪酸组成研究 被引量:1

摘要 采用松花粉诱导法从海藻、腐木等海洋漂浮物上分离得到6株破囊壶菌,其中菌株PC003、PC021、PC022为Schizochytrium属,菌株PC004、PC014、PC060为Thraustochytrium属.经分析,分属2个属的菌株的脂肪酸差异较大,Schizochytrium属的3个菌株含有占总脂肪酸22%以上的二十二碳六烯酸(DHA),8.3%以上的二十碳五烯酸(EPA),9.4%以上的二十碳四烯酸(ARA)和19%以上的C16:0(软脂酸);Thraustochytrium属的3个菌株含有占总脂肪酸33%以上DHA,7.8%以上的EPA,和24.7%的C16:0.其中,Thraustochytrium属菌株PC004的脂肪酸中DHA含量高达56.32%,可培养成为DHA高产菌株.结合6株菌之间的亲缘关系推测,亲缘关系相近,所产脂肪酸种类及含量相似,因而从Thraustochytrium属中更容易筛选到DHA高产菌株. Six strains of thraustochytrids are isolated from organic materials floating on the ocean such as seaweed using pine pollen as bait. Strains PC003, PC021 and PC022 are identified asSchizochytrium. Their fatty acids are composed of more than 22% docosahexaenoic acid (DHA), 8.3% eicosapentaenoic acid (EPA), 9.4% arachidonic acid (ARA) and 19% C16:0. Strains PC004, PC014 and PC060 are identified asThraustochytrium. Their fatty acids are made up of more than 33% DHA, 7.8% EPA and 24.7% C16:0. Furthermore, the content of DHA in fatty acids of strain PC004 is about 56.32%, and it has the potential to be cultivated for potentially high DHA production. The analysis on the relationship between fatty acids compositions of these strains and their genetic has been made. It is speculated that they are closer in affinity, similar in composition and content of fatty acids. In conclusion, it is much easier to obtain high DHA yielding strains fromThraustochytrium.
