铁皮石斛茎非多糖与粗多糖体内外抗氧化活性的比较 被引量:17

Antioxidant activity comparison between non-polysaccharides and crude polysaccharides from Dendrobium officinale stems in vitro and in vivo

摘要 目的 :明确铁皮石斛茎非多糖成分的抗氧化活性并与粗多糖进行比较,为非多糖成分的开发利用提供理论依据。方法:水提醇沉法制备铁皮石斛茎粗多糖,85%乙醇提取液合并醇沉上清液经制备后得到非多糖。以VC为阳性对照,采用ABTS法、DPPH自由基清除法及PMS-NBT-NADH法分别研究非多糖与粗多糖对ABTS自由基、DPPH自由基及超氧阴离子(O2·-)的清除能力。采用高糖高脂复合饮酒制备氧化损伤大鼠模型,灌胃给予铁皮石斛茎总提取物、非多糖、粗多糖(剂量均为1、2 g/kg,按生药计),6周后测定大鼠血清T-AOC、SOD及MDA。结果:在ABTS自由基、DPPH自由基、O2·-清除能力方面,非多糖优于粗多糖(非多糖的半抑制浓度IC50分别为0.357、1.545、3.535 mg/ml,粗多糖的IC50分别为6.597、2.077、4.916 mg/ml)。铁皮石斛茎总提取物、非多糖、粗多糖均能明显升高模型大鼠血清T-AOC值及SOD活力,降低MDA含量。结论:铁皮石斛茎非多糖成分在体内外均具有较强的抗氧化活性,并在清除ABTS自由基方面优于粗多糖,值得进一步开发并加以利用。 Objective: To compare the antioxidant activities of non-polysaccharides and crude polysaccharides from Dendrobium officinale stems in vitro and in vivo. Methods: Crude polysaccharides from Dendrobium stems was extracted with water and then continuously deposited by ethanol,non-polysaccharides was extracted with 85% ethanol. Their antioxidant activities were evaluated by various systems in vitro,including scavenging activity of superoxide anion,ABTS and DPPH radicals with Vc as positive control. Feed the rats with high glucose and high fat diet,and drink alcohol. Total extract,non-polysaccharides and crude polysaccharides( dosages were 1,2 g / kg) were administered for 6 weeks,then measured T-AOC,SOD,MDA on serum. Results: Both samples showed inhibitory effects on superoxide,ABTS and DPPH radical( IC50 of non-polysaccharide was 3. 535,0. 357 and 1. 545 mg / ml while 4. 916,6. 597 and 2. 077 mg / ml with polysaccharides). All of extractives could increase T-AOC and SOD( P 〈 0. 01,P 〈 0. 05),reduce MDA( P 〈 0. 05). Conclusion: Non-polysaccharide showed strong antioxidant activity in vitro and in vivo. In ABTS,non-polysaccharide showed stronger antioxidant activity,which worth further developing and utilizing.
