摘要
目的:通过观察荜茇根对实验性胆汁反流性胃炎(bile reflux gastritis,BRG)模型大鼠胃黏膜病理改变、血清胃泌素(Gastrin,GAS)水平、胃黏膜p21、PCNA、C-erb B-2蛋白表达情况的影响,探讨不同荜茇根有机溶剂提取物对胆汁反流性胃炎的药理作用及机制。方法:将140只SD大鼠随机分为正常组,模型组,多潘立酮组3mg/kg,乙醇提取物125mg/kg、250 mg/kg、400mg/kg剂量组,二氯甲烷提取物125mg/kg、400mg/kg剂量组,乙酸乙酯提取物125mg/kg、250 mg/kg、400mg/kg剂量组,正丁醇提取物125mg/kg、250 mg/kg、400mg/kg剂量组,观察各组大鼠胃黏膜病理表现,放射免疫法测定血清GAS的水平,免疫组化法测定胃黏膜p21,PCNA,C-erb B-2的蛋白表达量。结果:与模型组相比,多潘立酮组、荜茇根不同提取物组大鼠胃窦组织炎症有所减轻,血清GAS与p21蛋白表达水平也有不同程度的升高,差异有统计学意义(P<0.05)。结论:荜茇根不同提取物,尤其是乙醇和二氯甲烷提取物可以明显改善胆汁反流性胃炎模型大鼠胃黏膜炎症程度,可能与其部分成分可提高血清GAS水平有关,并通过对胃黏膜p21蛋白高表达进一步抑制炎症慢性进展。
Objective: Through the observation of different extracts of radix Piperis longi of experimental bile reflux gastritis( BRG) model in rat gastric mucosa pathological change,the level of serum gastrin( GAS),gastric mucosal p21,PCNA,C-erb B-2 protein expression of different Piper longum organic solvent extracts on bile reflux gastritis of pharmacological effects and its mechanism. Methods: 140 SD rats were randomly assigned to normal group,model group,domperidone group,extract of ethanol,low,middle and high dose group,extract of dichloromethane,low and high dose group,extract of Ethyl acetate,low,middle and high dose group,extract of N butyl alcohol,low,middle and high dose group. Observe the pathological features of gastric mucosa,the level of serum GAS are measured by radioimmunoassay. The expression of P21 in gastric mucosa is determined by immunohistochemical method. Results: Compared with the model group,Radix Piperis longi extract reduced inflammation of gastric antrum tissue,increased the serum of gastrin and p21 protein expression in different degree,the difference was statistically significant( P 〈 0. 05). Conclusion: Radix Piperis longi can improve bile reflux gastritis model rats gastric mucosa inflammation and may be related to the increase of serum GAS level and p21 protein expression in gastric mucosa to further inhibit the progress of chronic inflammation.
