摘要
目的研究参麦注射液对兔膝骨关节炎模型的干预作用,并探讨其作用机理。方法行前交叉韧带切除法(ACLT)复制兔膝骨关节炎模型,予膝关节腔注射参麦注射液、透明质酸钠4周后,取膝关节液及关节软骨组织,用ELISA法检测关节液白介素-1β(IL-1β)、白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)水平,RT-q PCR检测关节软骨组织IL-1β、IL-6、TNF-α基因表达。结果与正常对照组比较,模型组关节液IL-1β、IL-6和TNF-α的表达均明显升高(P<0.01),软骨组织IL-1β、IL-6和TNF-α的mRNA表达均明显升高(P<0.01);与模型组比较,参麦注射液组和透明质酸钠组关节液IL-1β、IL-6和TNF-α表达均显著降低(P<0.01),软骨组织IL-1β、IL-6和TNF-αmRNA均显著降低(P<0.05)。参麦注射液组与透明质酸钠组各指标比较差异无统计学意义(P>0.05)。结论参麦注射液对兔膝关节软骨有一定的保护作用,其作用机制与下调关节液IL-1β、IL-6、TNF-α水平,降低软骨组织IL-1β、IL-6、TNF-αmRNA表达有关。
Objective To study the therapeutic effect of shenmai injection in rabbits with knee osteoarthritis （KOA） and its potential mechanism. Methods KOA was induced in rabbits by anterior crueiate ligament transection （ ACLT）. Then shenmai and hyaluronie acid were administrated by intra-articula injection. After four weeks, the rabbits were killed and synovial fluid and cartilage were collected. The levels of IL-1β, IL-6 and TNF-α in synovial fluid were detected by ELISA. The expression of IL-1β, IL-6 and TNF-α mRNA in cartilage was detected by real-time quantitative PCR. Results Compared with the normal control group, the expressions of IL-1β, IL-6 and TNF-α in synovial fluid and cartilage were significantly increased in the model group （P〈0.01）. However, the expressions of these cytokines in synovial fluid and cartilage wereobviously decreased in model mice when treated with shenmai and hyaluronic acid （ P〈0.05）. There was no statistical difference between shenmai group and hyaluronic acid group （P〉 0.05）. Conclusion Shenmai injection has a protective effect on rabbit knee articular cartilage, which may be related to down-regulation of the expression of IL-1β, IL-6 and TNF-α in synovial fluid and cartilage.
