摘要
为筛选获得甜菜耐盐相关蛋白、探讨甜菜的耐盐机理以及培育耐盐甜菜品种,采用TCA丙酮沉淀法提取经高盐(300mmoL/LNaCl)处理组和对照组的甜菜叶片蛋白,利用双向电泳对蛋白进行分离,经考马斯亮蓝染色后获得分辨率高、重复性较好的双向电泳图谱,扫描后借助软件ImageMaster2DPlatinum5.0分析蛋白质图谱。实验结果表明:共找出47个差异蛋白点,进一步分析比对,鉴定出7种,这些蛋白多数与呼吸、能量代谢及光合作用有关。
To obtain salt tolerance related proteins, exploring the mechanism of salt tolerance and cultivated salt - tolerant beet varieties ; we extracted the beet leaf protein in high salt (300 mmol/L NaC1) treatment group and control group by TCA acetone precipitation method, isolated these protein by two - dimensional electrophoresis and coomassie brilliant blue staining; after that we got the high resolution, good repeatability .two - dimensional electrophoretogram and analysis the protein map by ImageMaster 2 d Platinum 5.0. The results showed that 47 differential protein were identified and further analysis confirm 7 proteins related to respiration, energy metabo- lism and photosynthesis.
