摘要
[目的]探索龙眼胚性愈伤组织中多糖积累最适宜的LED光质。[方法]以龙眼胚性愈伤组织为材料,采用单因素分析方法研究不同培养时间和LED光质对其生长和多糖积累的影响。[结果]研究结果表明,龙眼胚性愈伤组织在MS+2,4-D 1.0 mg/L培养基上暗培养15 d时多糖含量最高,为6.93%,SPSS分析表明,不同天数之间多糖含量存在显著性差异;在不同的光质处理下,多糖含量从高到低依次为:LED白光(15.22%)>LED红光(13.61%)>LED橙光(13.35%)>LED绿光(13.19%)>LED暖白光(8.00%)>LED蓝光(6.55%),以黑暗条件为对照,LED白光、暖白光和蓝光具有显著性差异,而LED红光、绿光和橙光不具有显著性差异;此外,黑暗条件最有利于龙眼胚性愈伤组织生长,LED白光和暖白光次之,而LED绿光和蓝光下愈伤组织生长状态最差。[结论]研究认为,LED白光培养15 d最有利于龙眼胚性愈伤组织多糖的积累,而LED暖白光和蓝光对多糖积累具有抑制作用,LED红光、绿光和橙光则作用不明显。试验结果为后期龙眼胚性愈伤组织多糖分子机制研究提供了科学依据。
[Objective] The aim was to explore the most appropriate LED light for accumulating polysaccharide in embryogenic callus of Dimocarpus longan Lour.(longan). [Method] In this experiment,embryogenic callus of Dimocarpus longan Lour.(longan) were used to study the effects of time and LED light on growth and polysaccharides accumulation of them. [Result] The results showed that, the polysaccharide content got the highest at 15 d, with the number of 6.93%,. SPSS analysis indicated that there were significant differences bewteen different times. In different LED lights, the accumulation of polysaccharides was LED white(15.22%) 〉LED red(13.61%)〉 LED orange(13.35%) 〉 LED green(13.19%) LED warm white(8.00%) LED blue(6.55%), and there were significant differences in LED white, warm white and blue when compared with it was in dark, while there were not in others. In addition, longan embryogenic callus grew best in dark, and second in LED white and warm white, when got the worse results in LED green and blue.[Conclusion] Our research suggested that, the optimum system for polysaccharide accumulation in longan embryogenic callus was LED white with a culture time of 15 d. The LED white could contribute to the accumulation of polysaccharides while LED blue and warm white would inhibit it. This study would provide scientific basis for further research on molecular-mechansim of polysaccharide in longan.
