针刺对DMBA诱导乳腺癌大鼠预防作用 被引量:4

Preventive Effect of Acupuncture in Rats with DMBA Induced Mammary Cancer

摘要 目的:探讨针刺足三里、三阴交、膻中对二甲基苯蒽(7,12-demethylbenz[a]anthrancene,DMBA)诱导的乳腺癌大鼠的预防作用,探讨针灸预防乳腺癌的有效性、针对性和可操作性。方法:将120只健康纯种的SD雌性6~8周龄大鼠随机分为模型A组(40只)、模型B组(40只)、空白组(40只),模型A、B组分2次灌注DMBA混悬液(10 mg DMBA/1 m L芝麻油)100 mg/kg体重,,每次灌胃50 mg/kg,间隔1周,复制大鼠乳腺癌病变模型;空白组灌服等容积芝麻油;模型A组于第2次灌胃后给予针刺治疗,针刺穴位为足三里(双侧)、三阴交(双侧)、膻中。观察DMBA对大鼠乳腺的影响,记录乳房外观、发育情况,手触大鼠各对乳房;采用B超检测除肉眼可见肿瘤的大鼠,记录大鼠乳腺表层至脏层的厚度;用精密度为1 mm的游标卡尺测量第2对、第3对乳房直径;Olympus光学显微镜下观察肿瘤组织病理学变化。结果:与空白组比较,模型A、B组大鼠成癌时间早、肿瘤发生率高,在B超检测下皮肤表层至脏层均有不同程度的增厚,大鼠乳头高度及直径也有不同程度的增高增宽,与模型A组比较,模型B组表现的更为显著(P〈0.05)。结论:针刺足三里、三阴交、膻中能减少DMBA诱导的大鼠发生乳腺癌的概率及时间,降低大鼠因DMBA毒性反应造成的死亡率,能够减缓DMBA诱导的乳腺癌大鼠"正常-一般增生-非典型增生-原位癌-浸润性癌"的演变过程。说明针刺对DMBA诱导的大鼠乳腺癌具有预防作用,其作用机制可能与调节机体性激素受体水平及其信号传导、提高机体免疫能力和调控人体基因表达有关。 Objective:To evaluate the preventive effect of aeupuncturing at Zusangli, Sangyingjiao and Danzhong in rats with dimethylbenzanthracine ( DMBA )-induced by mammrary cancer, to explore the effectiveness, maneuverability and pertinence of acupuncture prevent mammary cancer. Methods : 120 female SD rats aged 6-8 weeks were randomized into a model group A of 40 rats, a model group B of 40 rats and a blank group of 40 rats, the model group A, B received an oral gavage of DMBA ( 10 mg DMBA/1 mL sesame oil ) for model making. The blank group received an oral gavage of equal volume of sesame oil.The model of group was acupunctured affter second lavage, acupuncture at Zusangli, Sanyingjiao, Danzhong; and the model of group A and blank group, only the same grasp and release. To observe DMBA effects on mammary gland of rats, record the mammary appearance and development situation, and touch the breast in rats ; To use B-mode uhrasonograph addition to the visible to the naked eye tumors in rats, and record the rat mammary gland thickness of top layer to dirty, and measured with a precision of 1 mm of vernier caliper for 2,3 mammary diameter, to observe mammary tumor tissue pathological changes with push around optical microscope.Results : Compared with the blank group, model group rats into cancer occurred earlier, tumor incidence rate is high, the B-mode ultrasonograph is used to inspect the skin surface layer to dirty all have different degrees of thickening, nipple height and diameter of rats increased also have varying degrees of broadening ; compared with the model group A, the model B group showed more significant ( P〈O.05 ) . Conclusion: Acupunctureing Zusangli, Sanyingjiao, Danzhong can reduce the probability and time in rats with DMBA induced mammary cancer, and reduce the mortality of rats caused by DMBA toxic effects, and slow clown the "normal - general - atypical hyperplasia-carcinoma in situ and invasive carcinoma" belongs to the evolution process in rats with DMBA induced mammary cancer, that acupuncture has prevention function in rats with DMBA induced mammary cancer, its mechanism may be related to regulate the body's level of sex hormone receptor and its signal transduetion, and improve the body's immune ability and regulate gene expression related to human body.
