摘要
目的:研究健脾化湿解毒方含药血清对尿酸刺激下大鼠肾小球系膜细胞的增殖、纤维化相关因子表达的影响。方法:1将肾小球系膜细胞株(GMC)加入含有UA300μmol/L的培养液中;2分别加入含有别嘌醇(西药对照组)及健脾化湿解毒方的含药血清(中药治疗组)于细胞中;3用Cell Counting Kit观察各组别细胞的活性;用Western Blot检测细胞蛋白中磷酸化细胞外蛋白调节激酶(p-ERK)的含量;用共聚焦及Real-Time PCR检测细胞中纤维粘连蛋白(FN)的表达及分布情况,观察健脾化湿解毒方对于尿酸刺激下肾小球系膜细胞的增殖、纤维化是否具有保护作用。结果:尿酸300μmol/L刺激GMC后,细胞活性(CCK8)明显下降,p-ERK通路表达明显升高,细胞中FN的表达升高;在加入含别嘌醇及健脾化湿解毒方的含药血清后,细胞活性(CCK8)被进一步遏制;p-ERK通路与空白对照组相比并无明显差异。结论:健脾化湿解毒方的含药血清能降低经尿酸刺激后FN的分泌。
Objective: To study the effcacy of serum containing the Jianpi Huashi Jiedu decoction on GMC proliferation and ifbrosis related factors of rats under the stimulation of uric acid. Methods: First, GMCs were stimulated by UA (300μmol/L). Second, the cells in the western medicine control group were added into allopurinol; the cells in the TCM treatment group were added into serum containing the Jianpi Huashi Jiedu decoction. The activity of cell was detected by Cell Counting Kit. P-ERK in cell protein was detected by Western Blot. The expression and location of FN in cells were detected by confocal and Real-Time PCR. The protective effcacy of the Jianpi Huashi Jiedu decoction on GMC proliferation and ifbrosis of rats under the stimulation of uric acid was observed. Results: GMCs were stimulated by UA (300μmol/L), the activity of cell was decreased signiifcantly; p-ERK pathway expression was increased signiifcantly; expression of FN in cells was elevated. After added serum of serum containing allopurinol and the Jianpi Huashi Jiedu decoction, the activity of cell was further curbed; the differences of p-ERK pathway among three groups were not obvious. Conclusion: Serum containing the Jianpi Huashi Jiedu decoction can reduce the secretion of FN under the stimulation of uric acid.
