摘要
为给冬枣(Ziziphus jujube Mill.)黑斑病的生物防治提供优良菌株资源,以冬枣黑斑病菌为靶标,从冬枣健果中进行拮抗微生物资源的筛选。利用梯度稀释法从健康冬枣果实表面分离出细菌397株,其中K5-4对冬枣黑斑病菌的抑菌效果最为明显,抑菌带宽度为10.06 mm,该菌的无菌发酵液也具有良好的抑菌活性。活体防病试验发现,该菌能够有效地抑制冬枣黑斑病的发生,浓度为1×108cfu/m L时对黑斑病的防效为78.50%。该菌能够在冬枣果实中定殖,在接种后48 h内K5-4菌数增长为初始值的31.62倍,有无黑斑病病原菌存在对K5-4的定殖影响不大。通过形态学观察、生理生化试验以及16S r DNA序列同源性分析,鉴定该菌株为解淀粉芽孢杆菌(Bacillus amyloliquefaciens),其在防治冬枣黑斑病上有较好的应用前景。
To provide excellent biocontrol strain resources against Dongzao( Ziziphus jujube Mill.) black spot disease,with the pathogen Alternaria alternate as target,antagonistic microbial resources were screened from health jujube fruit. A total of 397 bacteria were isolated and purified from healthy jujube fruit surface using dilution method. Strain K5-4 had the most obvious effect with 10. 06 mm inhibition zone. Its sterile broth had a good antibacterial activity also. Treatment with 1 × 108 cfu / m L cell suspension of K5-4 could control the black spot disease on Dongzao of 78. 50%. K5-4 could colonize in Dongzao fruit with or without the presence of black spot pathogen,and within 48 h after inoculation the number of K5-4was 31. 62 times the initial value. By morphological observation,physiological and biochemical characterization and the analysis of the 16 S r DNA sequence,this strain was identified as Bacillus amyloliquefaciens. The strain K5-4 has a good prospect in controlling black spot disease on jujube fruit.
