摘要
采用超高效液相色谱-串联质谱(UPLC-MS/MS)法检测自制药酒中乌头碱、次乌头碱和中乌头碱等三种乌头类生物碱的含量。样品过滤后,经电喷雾电离,采用多反应监测模式进行检测,外标法定量。结果表明,添加浓度为5-100ng/mL时,乌头类生物碱的回收率为86.7-106.2%,相对标准偏差为1.4-5.7%。方法简便快捷,精密度高,重现性好,可用于自制药酒中乌头类生物碱含量的测定。
An ultra performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS)method was estab- lished to determine aeonitum alkaloids, including aconitine, hypaconitine, mesac0nitine in herb wine. Samples were filtrated directly, and then UPLC-MS/MS system coupled with an electrospray ionization (ESI) source was performed using multiple reaction monitoring modle. The quantification was obtained utilizing the external standards. Recoveries of the alkaloids ranged from 86.7% to 106.2% at fortified levels of 5-100ng/mL, the RSDs of the method were in the range of 1.4-5.7%. The method is simple, effective and accurate. It can be applied to determine the alkaloids in herb wine simultaneously.
