The correlation between plasma homocysteine and contents of hydrogen sulfide in the corpus cavernosum of hyperhomocysteinemia in rats

摘要 目的:研究高同型半胱氨酸血症(Hhcy)大鼠血浆同型半胱氨酸(Hcy)浓度和阴茎海绵体组织硫化氢(H_2S)含量之间的关系。方法:将24只健康雄性Wistar大鼠适应喂养1周后随机分为8只正常组(C组)和16只模型组,给予C组(生理盐水3m L/次)和模型组(生理盐水3m L/次+蛋氨酸1g·kg^(-1)·d^(-1))早晚各1次灌胃,持续6周。6周后采血测模型组血浆Hcy浓度均大于16.0μmol/L,提示造模成功,随机将模型组分为Hhcy组(H组)和叶酸、vit B_(12)组(V组)各8只,给予H组(生理盐水3m L/次+蛋氨酸1g·kg^(-1)·d^(-1))和V组(生理盐水3m L/次+蛋氨酸1g·kg^(-1)·d^(-1)+叶酸0.5mg·kg^(-1)·d^(-1)+vit B_(12)50μg·kg^(-1)·d^(-1))继续灌胃6周后测血浆Hcy和H_2S浓度,0、5V下阴茎海绵体内压/平均颈动脉压(ICPmax/MAP)检测后取阴茎组织测H_2S含量。结果:随着C组、V组、H组之间血浆Hcy浓度依次增高,而大鼠ICPmax/MAP(0、5V)值逐渐降低,阴茎组织H_2S含量依次均显著性降低(P<0.05),且血浆Hcy浓度与阴茎组织H_2S含量之间为负性相关。结论:随着血浆Hcy浓度增高,致使阴茎海绵体内源性H_2S含量降低,这可能是Hhcy致勃起功能下降的原因之一,且叶酸和维生素B_(12)能够有效的降低血浆Hcy浓度,改善勃起功能。 Objective: To study the relationship between plasma homocysteine( Hcy) and contents of hydrogen sulfide( H_2S) in the corpus cavernosum of hyperhomocysteinemia( Hhcy) rats; and to explore the mechanism of Hhcy rats with decreased erectile function. Methods: Twenty four healthy male wistar rats were fed in the same conditions. One week later,they were randomly divided into control group( C group with eight rats) and model group( sixteen rats). By intragastric administration and twice one day,model group rats were given with methionine( 1g·kg^-1·d^-1) and physiological saline( 3m L per time) while group C with physiological saline( 3m L per time). Six weeks later,the levels of model group rats were all over 16. 0 μmol/L. Then model group rats were randomly divided into Hhcy group( H group,eight rats) and vitamin B12group( V group,eight rats). The group H was given with methionine and physiological saline by intragastric administration and twice one day like before while group V was given with methionine( 1g·kg^-1·d^-1) and physiological saline( 3m L per time) and folate( 0. 5mg·kg^-1·d^-1) and vitamin B12( 50μg·kg^-1·d^-1). Six weeks later,all rats were detected the concentrations of Hcy in plasma and H2 S in the plasma and penile tissue. Results: As the levels of plasma Hcy of group C、V and H became higher and higher,the contents of H2 S in the plasma and penile tissue correspondingly became lower and lower. There was negative correlation between plasma Hcy and H2 S in the corpus cavernosum. Conclusion: Increased levels of plasma Hcy may result in reduction of endogenous H2 S,and this may be the reason of reduced erectile function with Hhcy. Folate and vitamin B12 could make erectile function better.
