摘要
以从中温大曲中筛选获得的紫色红曲霉FBKL3.0018为研究对象,并对该菌株液态发酵产酯化酶工艺条件进行优化。通过单因素试验、正交试验确定此菌株优化后的液态发酵的最佳培养基组成为5%麦芽糖、2%牛肉膏、0.04%硫酸镁,最高酶活力达到116.32 U/m L,较培养基优化前提高45%;最佳培养条件为接种量10%、培养温度32℃、培养时间5 d、摇床转速160 r/min,最高酶活力达到153.34 U/m L,较培养条件优化前提高32%。
Using the Monascus purpureus FBKL3.0018 screened from medium temperature Daqu as research object, the esterification enzyme-produc- ing technology conditions of submerged fermentation were optimized. By single factor experiments and orthogonal experiments, the optimum com- ponents of culture medium were determined as maltose 5%, beef extract 2% and MgSO40.04%, and the highest enzyme activity was up to 116.32 U/ml, which was 45% higher than that of before optimization. The optimum culture conditions were inoculum 10%, culture temperature 32 ℃, time 5 d and shaker speed 160 r/min, and the highest enzyme activity was up to 153.34 U/ml, which was 32% higher than that of before optimization.
