摘要 目的:针对中药制剂湿润烧伤膏,建立其中罂粟壳所含罂粟碱、吗啡、那可丁、可待因、蒂巴因等5种化学成分的凝胶渗透色谱-超高效液相色谱-质谱/质谱联用的检测方法。方法:色谱柱为Phenomenex Kinetex HILIC 100A(100 mm×2.1 mm,1.7μm);柱温为40℃;流动相为10 mmol/L甲酸铵(以甲酸调节p H值至3.0)及乙腈,梯度洗脱;流速为0.2 m L/min,以优化后的质谱参数采集信号。结果:各成分检出限浓度为0.1~0.5 ng/m L;定量限浓度为0.5~1.0 ng/m L;线性范围为各成分定量限~100 ng/m L;在线性范围内相关系数均在0.99以上;平均加样回收率86.5%~99.6%。结论:本方法准确,灵敏度高,专属性好;能用于湿润烧伤膏中罂粟壳化学成分的检测。 Objective :To develop a GPC-UPLC-MS/MS method for determination of five chemical components( papaverine,morphine,narcotine,codeine and thebaine) of papaveris pericarpium in MEBO.Methods : Phenomenex Kinetex HILIC 100 A column(100 mm × 2.1 mm,1.7 μm) was adopted.The column temperature was40 ℃.The mobile phase was 10 mmol/L ammonium formate(p H 3.0)-acetonitrile,which was used for gradient elution at a flow rate of 0.2 m L/min.The signals were collected by using the optimized parameters.Results : The detection limit of this method was 0.1 ~ 0.5 ng/m L,while the limit of quantitation was 0.5 ~ 1.0 ng/m L,and the linear range was the limit of quantitation ~ 100 ng/m L.The correlation coefficients within the linear range were more than 0.99.The average recoveries of test samples were 86.5% ~ 99.6%.Conclusion :This method shows high accuracy,sensitivity and specificity,which may be used for the determination of five chemical components of papaveris pericarpium in MEBO.
