摘要
目的采用非暴露式气管注入方法制备大鼠矽肺模型。方法取SPF级SD大鼠48只,24只由大鼠肺部注入50 mg/只的二氧化硅粉尘,24只注入1 ml/只生理盐水,然后连续30 d超声波喷雾生理盐水。处死动物后通过测量脏体系数、全肺胶原含量、干湿重、游离二氧化硅含量、肺灌洗液中细胞计数及分类,观察肺组织病理学变化,全面评估大鼠矽肺模型制备效果。结果模型组大鼠脾、肺、肾脏器系数、肺湿重、干重、细胞总数、中性粒细胞率、羟脯氨酸含量分别为0.252%、1.294%、0.729%、3.045 g、1.090 g、0.833×109个/ml、75.083%、0.728 mg/g,明显高于阴性对照组,差异有统计学意义(P<0.05);淋巴细胞、巨噬细胞率分别为11.916%、13.000%,明显低于阴性对照组,差异有统计学意义(P<0.05);肺组织可见散在或灶性纤维细胞结节,部分结节胶元纤维形成并相互融合连成片,形成结节为主的结节性肺纤维化,使肺组织完全实变。结论通过非暴露式气管注入石英粉尘,成功制备大鼠矽肺模型,为尘肺病的治疗研究提供途径。
[Objective]To establish the silicosis rat model by intratracheal administration of silica with non-exposure method.[Methods]Forty-eight SPF adult SD rats were randomly divided into the silicosis rat model group(n=24)and the control group(n=24). Rats were subjected to intratracheal administration of silica(50mg per rat)in the silicosis rat model group,while the control group was injected with the normal saline solution(1 ml per rat)and subsequently treated with ultrasonic spray of saline solution for 30 days. Afterwards,the animals were sacrificed and the silicosis rat model group was assessed comprehensively by measuring organ coefficient,collagen content,dry and wet weight and free silica of the lung,together with counting and classifying the cells of the lung lavage fluid,and observing the pulmonary pathological changes. [Results]The spleen coefficient,lung coefficient,kidney coefficient,wet weight of the lung,dry weight of the lung,the number of total cell count,percentage of theneutrophil phagocytosis,and the content of hydroxyproline in the silicosis rat model group was respectively 0.252%,1.294%,0.729%,3.045 g,1.090 g,0.833×109 /ml,75.083% and 0.728 mg/g,which were significantly higher than those in the control group(P〈0.05). The percentage of lymphocytes and macrophages in the model group was 11.916% and 13.000% respectively,which were significantly lower than those in the control group(P〈0.05). The scattered or focal fibred cell nodules were observed in the silicosis rat model group,and part of the nodular rubber fiber produced and fused with each other and formed nodular pulmonary fibrosis.[Conclusion]The silicosis rat model is established successfully by intratracheal administration of silica with non-exposure method,which would provide the foundation for the treatment of pneumoconiosis.
