摘要
目的研究2 940 nm Er∶YAG点阵激光照射后,对皮肤真皮结构和衰老机制的影响。方法将体质量(40±2)g健康雌性昆明小鼠72只随机均分为3组,分别标记为组1、组2、组3,每组24只。采取自身空白对照,记为组0。激光照射(18 J/cm2、24 J/cm2、30 J/cm2能量密度,对应组1、组2、组3)1、2、4周后分别处死组1、组2、组3小鼠各8只,取实验区域皮肤,取距离实验区域5 cm处背部皮肤为自身空白对照,进行丽春红-维多利亚蓝染色的病理组织观察,免疫组织化学染色观察金属基质蛋白酶-1(MMP-1)/金属蛋白酶组织抑制剂-1(TIMP-1)表达的改变。使用Image Pro Plus 6.0软件对图像进行定量分析,选择平均光密度值(mean OD)作为统计数据。结果胶原纤维/弹性纤维染色结果显示,胶原纤维和弹性纤维表达均明显增加(P<0.05),且不同剂量组间差异有统计学意义(P<0.05)。点阵激光照射1周后,MMP-1表达增加,其中组3(高能量组)更为明显(P<0.05)。2周后,MMP-1表达开始下降,4周后MMP-1表达显著下降(P<0.05),组3(高能量组)尤为明显(与组1、组2相比,P<0.05)。点阵激光照射1、2周后,TIMP-1表达均无明显改变(P>0.05),照射4周后可观察到TIMP-1表达上升,但差异无统计学意义(P>0.05),组间差异亦无统计学意义(P>0.05)。结论 2 940 nm Er∶YAG点阵激光照射短期内可造成小鼠真皮胶原纤维和弹性纤维变性,后期促进两者增生,提示点阵激光可用于瘢痕组织新生;短期内可增加小鼠真皮细胞内MMP-1的表达,后期可抑制其表达,提示点阵激光可用于皮肤抗衰老治疗。
Objective To study the effect of 2 940 nm Er : YAG laser on skin dermis and aging mechanism, and provide theoretical basis for its clinical application. Method Seventy-two healthy female Kunming mouse with weight of(40 ± 2) g were randomly divided into 3 groups and marked as group 1, group 2 and group 3(n = 24), the self blank control recorded as group 0.Eight mice in each group were sacrificed 1-week, 2-week and 4-week after laser irradiation(18 J/cm^2, 24 J/cm^2, 30 J/cm^2), respectively. The skin of experimental area was obtained and the back skin 5 cm away from experimental region was set as blank control,and then Ponceau S-Victoria blue staining was performed to observe the collagen fiber/ elastic fiber morphology and immunohis-tochemical staining was conducted to detect the expression of MMP-1/ TIMP-1. The Image Pro Plus 6.0 software was used for quantitative analysis of the image, with mean optical density value(mean OD) as statistical data. Results The collagen/ elastic fiber staining showed that collagen fiber and elastic fiber expression were increased significantly after irradiation( P〈0.05), and there were statistically significant differences among the different dosage groups(P〈0.05). One-week after dot matrix laser irradiation, the expression of MMP-1 was increased and that of group 3 was more significant(P〈0.05). At 2-weeks, the expression of MMP-1 began to decline. At 4-week, the expression of MMP-1 was significantly decreased( P〈0.05), and that of group 3 was decreased more obviously( P〈0.05) as compared with that of group 1, group 2. One-week and 2-week after dot matrix laser irradiation, expression of TIMP-1 was not obviously changed(P〉0.05). After 4-week, TIMP-1 expression was increased but without statistical significance(P〈0.05), and there was no significant difference among different groups(P〉0.05).Conclusion It is demonstrated that 2 940 nm Er ∶ YAG laser irradiation caused degeneration of mouse dermal collagen fibers and elastic fibers in the earlier stage, but later promoted hyperplasia of both, which is positively correlated with laser energy,suggesting that dot matrix laser could be used for skin anti-aging treatment.
