摘要 目的:观察阿魏酸对体外培养大鼠成骨细胞增殖、碱性磷酸酶(ALP)、骨保护素(OPG)及骨活素(OA)表达的影响,探讨阿魏酸在骨折后骨愈合中的作用。方法:以培养基为空白对照,MTT法检测阿魏酸200、20、2μg/L培养后0、1、2、3、4、5、6 d成骨细胞增殖的变化,给药后6 d,ELISA法检测细胞上清液ALP活性,RT-PCR检测OPG mRNA和OA mRNA表达的变化。结果:阿魏酸可剂量依赖性促进体外培养大鼠成骨细胞的增殖,显著性增加ALP的活性、OPG mRNA和OA mRNA的表达(P<0.05)。结论:阿魏酸具有骨形成促进效应,其机制可能与其促进成骨细胞增殖与分化、改善骨代谢有关,适用于临床骨折的辅助治疗。 Objective: To observe the effects of Ferulic Acid on proliferation of rat's osteblast and its alkaline phosphatase activity,osteoprotegerin( OPG) and osteoactivin( OA) gene expressions in vitro,explore the effect of Ferulic Acid in fracture healing. Methods : Cultured rat's osteoblasts were added into 200,20,2 and 0 μg / LFerulic Acid respectively,proliferation variation tendency of osteoblasts in cultured for 0 - 6 days was measured by MTT. Alkaline phosphatase activity of osteoblasts was determined by ELISA,OPGmRNA and OAmRNA expressions were determined by RT- PCR in the sixth day. Results: Ferulic Acid could accelerate the proliferation of osteoblasts in a dose- dependent manner,and also enhance the intracellular alkaline phosphatase activity and OPG and OA gene expressions significantly.( P〈0. 05). Conclusion: Ferulic Acid has effect of promoting bone formation,the mechanism is related to accelerate the proliferation and differentiation of osteoblast and improve the bone metabolism.
