摘要
目的:研究运动训练对2型糖尿病大鼠肾脏NF-κB-p65及相关蛋白表达的影响,以探讨运动改善2型糖尿病大鼠肾损伤的可能作用机制。方法:SD大鼠通过高脂饲料喂养6周和低剂量腹腔注射STZ的方法复制2型糖尿病大鼠实验模型。实验分为正常对照组(A),糖尿病安静组(B)和糖尿病运动组(C)。C组进行6周的有氧游泳训练,其余两组不施加干预。实验结束后,测试随机血糖、24 h UAE、T-SOD、MDA、NF-κB-p65、Activated Caspase-3和Nephrin蛋白表达,并进行电镜观察肾脏显微结构的改变。结果:1)电镜下,运动干预的C组大鼠可见滤过膜3层结构较清晰,基底膜未见明显增加,足突稍肿大、足细胞融合等症状较B组大鼠均有所改善。2)运动干预后,C组大鼠的血糖浓度、24h UAE排泄量均降低,与B组比较,均有显著性差异(P<0.05或P<0.01)。3)运动干预后,C组大鼠肾皮质T-SOD活性上升而MDA含量下降,与B组比较,亦有明显差异(P<0.05或P<0.01)。4)运动干预后,C组大鼠肾皮质NF-κB-p65和Activated Caspase-3蛋白表达均明显降低,而Nephrin蛋白表达明显增加,与B组比较,差异均呈显著性(P<0.05或P<0.01)。结论:运动能降低2型糖尿病大鼠血糖,减轻肾脏病理损伤,并提高肾功能,其机制可能与运动的下调氧化应激介导的2型糖尿病大鼠肾脏核转录因子NF-κB-p65的表达,以改善效应蛋白Activated Caspase-3异常表达和恢复足细胞特异标志物-Nephrin蛋白的表达,而改善大鼠肾脏损伤的保护作用有关。
Objective:To investigate the effect of exercise training on NF-κB-p65 and related proteins in type 2 diabetic rats and its mechanism.Methods:The model of type 2 diabetic rats was established through SD rats fed high-fat diet for six weeks together with intraperitoneal infecting after a low dose of STZ.The model rats were randomly divided into diabetic control group(B)and diabetic exercise group(C)with intervention of exercise for six weeks,additionally provided normal control group(A)without any intervention.We examined blood glucose concentrations and the excretion of 24 h microalbuminuria(UAE),and T-SOD activity and MDA content and the expressions of NF-κB-p65,Activated Caspase-3 and Nephrin in the renal cortex and glomerular ultrastructure changes were observed by electron microscope after the experiment end.Results:1)Group B could be found that it had glomerular wall oozing with the adhesions and part of telangiectasia,obvious glomerular basement membrane foot process fusion and so on,while the above glomerular injury improved significantly in Group C with exercise through electron microscope.2)Compared with Group B,the concentrations of blood glucose and 24 hUAE in Group C decreased significantly(P〈0.05 or P〈0.01).3)Compared with Group B,the activity of T-SOD in Group C increased significantly,while the content of MDA decreased significantly(P〈0.05 or P〈0.01).4)Compared with Group B,the expressions of NF-κB-p65 and Activated Caspase-3 significantly decreased,while Nephrin in Group C of the renal cortex significantly increased(P〈0.05 or P〈0.01).Conclusion:Exercise could improve blood glucose and renal function,and decrease renal injure for type 2 diabetes,which may be related to decreasing the protein over-expression of NF-κB-p65 induced by oxidative stress and Activated Caspase-3 activity,while increasing of Nephrin expression in type 2 diabetes after exercise.
