药用昆虫美洲大蠊内生放线菌的分离和鉴定 被引量:12

Isolation and identification of endophytic actinomycetes from medical insects(Periplaneta americana)

摘要 目的探讨药用昆虫美洲大蠊内生放线菌的多样性和生物学特性,挖掘新的微生物资源。方法采用平板稀释法对美洲大蠊内生菌进行分离和纯化;分离菌通过形态学观察和16SrDNA序列分析进行分类鉴定,运用MEGA5.1软件,采用最大似然值法构建系统发育树,对其进行进化分析。结果从15只美洲大蠊肠道分离得到159株内生放线菌,分属于12个属9个科,其中链霉菌107株,戈登氏菌13株,诺卡氏菌2株,小单孢菌5株,栖白蚁菌4株,纤维单孢菌3株,分枝杆菌4株,微杆菌11株,短状杆菌4株,无色杆菌4株,土壤泛球菌1株,壤霉菌1株。结论美洲大蠊的内生放线菌丰富多样,其中蕴藏着的新类型放线菌,可能成为活性天然产物和先导药物的重要来源。 Objective In order to mine new microbial resources, this study examined the diversity and biological characteristics of endophytic actinomycetes found in Periplaneta americana, a species of medical insects. Methods Plate dilution was used to isolate and purify endophytic actinomycetes from a sample of P. americana. The isolated strains of actinomycetes were first identified on the basis of morphological characteristics including the morphology of the spore chain, the color of aerial and substrate mycelium, and pigment production. Strains were then identified using 16S rDNA gene se- quencing. Phylogenetic analysis was done with software included in MEGA5.1 using the maximum likelihood method. Results One hundred and fifty-nine strains of endophytie actinomycetes were isolated from 15 P. americana. These strains belonged to 12 genera in 9 families and included 107 Streptomyces strains, 13 Gordonia strains, 2 Nocardia strains, 5 Micromonospora strains, 4 Isoptericola strains, 3 Cellulomonas strains, 4 Mycobacterium strains, 11 Microbacterium strains, 4 Leucobacter strains, 4 Achromobacter strains, 1 Terracoccus strain, and 1 Agromyces strain. Conclusion Various endophytic actinomycetes were found in P. americana, so these insects represent a new source of actinomycetes. These actinomycetes can facilitate the discovery of new active compounds.
