摘要
目的探究特异性蛋白3(Sp3)与三磷酸腺苷结合蛋白G亚族成员2(ABCG2)在人结肠癌组织中的表达及其相关性,并探讨其临床意义。方法收集2010年1月1日至2015年1月1日普通外科行外科手术治疗的50例患者的结肠癌组织及对应癌旁组织,采用免疫组化染色检测Sp3和ABCG2蛋白在组织中的表达,采用Spearman相关性分析两种蛋白在结肠癌组织中表达的相关性,分析肿瘤组织中Sp3和ABCG2蛋白表达的临床意义。结果Sp3和ABCG2蛋白均高表达于结肠癌组织中,其阳性表达率分别为50.0%和66.0%,明显高于癌旁组织中Sp3(20.0%)和ABCG2蛋白(26.0%)阳性表达率(P均<0.01)。临床资料分析发现,有淋巴结转移及TNM分期(Ⅲ+Ⅳ期)的结肠癌组织中Sp3及ABCG2蛋白阳性表达率明显高于无淋巴结转移和Ⅰ+Ⅱ期的结肠癌组织(P<0.01,P<0.05);相关性分析表明,Sp3和ABCG2蛋白在结肠癌组织中的表达呈显著正相关关系(r=0.801,P<0.01)。结论结肠癌组织中Sp3和ABCG2蛋白表达上调,且与患者恶性临床特征相关,二者表达的正相关关系提示在结肠癌中ABCG2可能是Sp3的转录靶点之一。
Objective To investigate correlation between expressions of specificity protein 3( Sp3) and ATP-binding cassette sub-family G member 2( ABCG2) in colon carcinoma tissues and their correlation and explore the clinical significance. Methods The colon carcinoma tissues and the matched tumor adjacent tissues samples from 50 patients receiving surgical operation between January 1,2010 and January 1,2015 were collected. Immunohistochemistry staining method was used to detect the expressions of Sp3 and ABCG2 proteins in the tissues. Spearman correlation analysis was used to analyze the relationship between these two proteins expressions in colon carcinoma tissues and the clinical significance of Sp3 and ABCG2 proteins expressions in colon carcinoma tissues. Results Both Sp3 and ABCG2 proteins were highly expressed in colon carcinoma tissues,and the positive expression rates of Sp3( 50. 0%) and ABCG2( 66. 0%) in colon carcinoma tissues were significantly higher than those( 20. 0%,26. 0%) in tumor adjacent tissues( all P〈0. 01). The clinical data analysis showed that the positive expression rates of Sp3 and ABCG2 proteins in colon carcinoma tissues with lymph node metastasis and TNM staging of III + IV stages were significantly higher than those without lymph node metastasis and with TNM staging of I + II stages( P〈0. 01,P〈0. 05). The correlation analysis showed that the expressions of Sp3 and ABCG2 proteins in colon carcinoma tissues presented significant positive correlation( r = 0. 801,P〈0. 01). Conclusions The expressions of Sp3 and ABCG2 proteins in colon carcinoma tissues are up-regulated and associated with patients' malignant clinic features. The positive correlation of Sp3 and ABCG2 expressions prompts that ABCG2 may be one of transcriptional targets of Sp3.
