摘要
目的研究贵州罗甸地区南方大斑蝥(Mylabris phalerata Pallas)和黄黑小斑蝥(Mylabris cichorii Linnaeus)体内的结合斑蝥素(conjugated cantharidin)对人肝癌Hep G2细胞增殖和凋亡的影响。方法采用两种斑蝥三氯甲烷提取物的水浸液获取结合斑蝥素。运用磺酰罗丹明染色法(SRB)检测结合斑蝥素对Hep G2细胞增殖的作用。利用Annexin V-FITC/PI双染法检测结合斑蝥素对Hep G2细胞凋亡的影响。应用倒置相差显微镜、透射电子显微镜观察凋亡细胞的形态学变化。结果两种结合斑蝥素对Hep G2细胞的增殖均有显著的抑制作用,呈剂量依赖性,其半数抑制浓度(IC50)分别为0.90、0.85 mg/L。流式细胞术结果显示,以0.90 mg/L的南方大斑蝥结合斑蝥素和0.85 mg/L黄黑小斑蝥结合斑蝥素作用Hep G2细胞24 h后,细胞呈典型的凋亡形态特征,凋亡率分别为(10.5±1.45)%和(12.8±1.53)%;结论两种来源斑蝥体内结合斑蝥素均可抑制肝癌Hep G2细胞的增殖并促进其凋亡。
AIM To investigate the influence of conjugated cantharidin from Mylabris phalerata Pallas ; Myla- bris cichorii Linnaeus, collected from Luodian District, Guizhou Province, on the proliferation and apoptosis of hu- man hepatoma HepG2 cells. METHODS Conjugated eantharidin was obtained from water soluble matter of tri- ehloromethane extracts of two sources of Mylabris. SRB assay was used to study the effects of conjugated cantharidin on the proliferation of HepG2 cells. Annexin V-FITC/PI staining was employed to detect the influence of conjugated cantharidin on the apoptosis of HepG2 cells. Morphological alteration of apoptotie cells was observed by phase con-trast microscopy and transmission electron microscopy, RESULTS Two kinds of conjugated cantharidin all exer- ted obvious inhibitory effects on the proliferation of HepG2 cells in a dose-dependent manner; the ICs0 value was 0. 90 mg/L and 0. 85 rag/L, repectively. The results of flow cytometry showed that represented ( 10. 5 ± 1.45 ) % and (12. 8 ± 1.53)% apoptosis rates, respectively, when 0. 90 mg/L conjugated cantharidin originated from M. phalerata and 0. 85 mg/L conjugated eantharidin originated from M. cichorii were incubated with HepG2 cells for 24 h. Morphological observation indicated that cells appeared typical characteristics of apoptotic morphology. CONCLUSION The conjugated eantharidin from two kinds of Malybris can inhibit the proliferation and induce the apoptosis of HepG2 cells.
