摘要
研究在不同骨关节炎中单克隆抗体(ED1)阳性细胞及降钙素基因相关肽(CGRP)阳性神经纤维在骨关节炎(OA)各个相同时期的表达变化及临床意义。将45只健康成年Wistar雄性大鼠随机分为正常组、手术组、跑步训练组,并进行OA造模。分别于1周、2周、3周、6周时处死大鼠,从各组对应时期取5只大鼠全膝关节组织置于4%PFA液固定、制片、免疫染色后,采用病理图像分析系统,通过观察及半定量ED1阳性细胞和CGRP阳性神经纤维的表达,评价两种骨关节炎中滑膜组织的早期病理变化。手术组及跑步训练组在1、2、3周较正常组均可见较多的ED1阳性细胞,手术组ED1阳性细胞升高明显,且手术组和跑步训练组在1、2、3周时ED1阳性细胞数呈极显著差异(P<0.01)。CGRP阳性神经纤维的表达显示:跑步训练组增多显著;2周时两组的CGRP阳性神经纤维数存在显著或极显著差异(P<0.05或0.01)。手术组的ED1阳性细胞的表达明显高于跑步训练组;而CGRP阳性神经纤维的表达显示跑步训练组高于手术组。
To investigate the expression of ED1 and calcitonin gene related peptide (CGRP) between the operation and running exercise for deferential osteoarthritis animal model, 45 male Wistar rats were randomly divided into 3 groups:control group (A), operation group (B) and running exercise group (C). The whole knee osteoarthritis of model rats were collected at 1, 2, 3 and 6 weeks and fixed in 4% PFA solution, section, immunohistochemical staining. The expression of ED1, CGRP and the pathological changes of the early osteoarthritis (OA) synovium were observed. Compared with group B, CGRP positive never fibers(7.753±0.736), (2.214±1.021) to group C was lighter (P〈0.05);ED1 positive cells expression(401.771±5.632), (353.440±4.321), (132.302±3.447)and(5.562±1.201), (18.665±1.533), (11.629±1.217)significantly higher (P〈0.01 or 0.05). ED1 positive cells expression in the group B is higher than that in group C;but Running exercise group.CGRP positive never fibers are higher than the operation group.
