摘要
目的研究燕麦膳食纤维(dietary fiber,DF)对高脂高胆固醇饮食小鼠肠道胆固醇吸收的影响及其机制。方法 36只雄性C57BL/6J小鼠,按体质量随机分为:阴性对照组、模型对照组、燕麦DF组,预防性干预24 w。实验结束后测定各组小鼠血清总胆固醇(TC)、甘油三酯(TG)及高密度脂蛋白胆固醇(HDL-C),计算低密度脂蛋白(LDL-c)。western blot方法测定肠道中与胆固醇吸收相关的GTP结合蛋白Sar1B、尼曼-匹克C1型类似蛋白1(NPC1L1)、清道夫受体B1(SR-B1)蛋白的表达水平。结果实验结束时,与模型对照组相比,燕麦DF组的体重、血清TC、LDL-C水平均显著下降(P<0.05),HDL-C水平显著升高(P<0.05);与胆固醇吸收相关的NPC1L1水平显著降低(P<0.05),SR-B1无显著变化,调控NPC1L1和SR-B1的Sar1B蛋白表达水平显著升高(P<0.05)。结论燕麦DF可以通过上调Sar1B降低肠道NPC1L1表达水平,从而减少肠道胆固醇吸收,降低血脂水平,改善高脂高胆固醇饮食引起的高脂血症。
Objective To explore the effects and mechanism of oat fiber on intestind cholesterol absorption in C57BL/6J mice fed a high-fat/cholesterol diet. Methods Thirty-six male C57BL/6J mice were randomly divided into three groups according to the body weight: negative control group, model control group, and oat fiber group. At the end of experiment, serum total cholesterol (TC), triglyceride (TG) and high-density lipoprotein cholesterol (HDL-C) were measured, and low-density lipoprotein cholesterol (LDL-C) was calculated. Protein expressions of GTP binding protein SarlB, niemann-pick C l-like 1 (NPC 1L 1) and scavenger receptorB 1 (SR-B 1) were analyzed by Western blot method. Results After 24 weeks of feeding, oat fiber significantly decreased body weight, serum TC and LDL-C concentrations, and increased serum HDL-C level compared with model control group (P〈0.05). Protein expression of SarlB was upregulated, and protein expression of NPC1L1 was downregulated in oat fiber group (P〈0.05). There was no significant statistical difference in protein expression of SR-B1 between the model control group and oat fiber group. Conclusion Oat fiber lowers intestind cholesterol absorption by activation of SarlB expression and suppression of NPC1L1 expression, and improves dyslipidemia induced by a high-fat/cholesterol diet.
