摩拉、尼里/拉菲种公牛促性腺激素释放激素受体(GnRHR)基因序列多态性检测及基因型分析 被引量:3

Detection of the sequence polymorphisms of GnRHR and its genotype analysis in Murrah and Nili-Ravi breeding bulls

摘要 为了分析摩拉、尼里/拉菲种公牛促性腺激素释放激素受体(GnRHR)基因的遗传多态性,试验对28头摩拉种公牛、40头尼里/拉菲种公牛采用PCR扩增、测序、高分辨率熔解曲线检测的方法对GnRHR基因进行遗传多态性检测。结果表明:摩拉、尼里/拉菲种公牛GnRHR基因存在相同的SNP位点,分别是A13 342G、T13 355G、A13 406T、T17 221C。其中前三个SNP位点位于第一内含子,最后一个SNP位点位于第三外显子,该突变位点使氨基酸编码由酪氨酸变为组氨酸。对于基因分型,摩拉、尼里/拉菲种公牛却出现不同结果,摩拉种公牛13 342位点有2种基因型AA、AG,频率分别为0.607 1,0.329 2;13 355位点有2种基因型TT、TG,频率分别为0.714 3,0.285 7;13 406位点有2种基因型AA、AT,频率分别为0.750 0,0.250 0;17 221位点有2种基因型TT、TC,频率分别为0.750 0,0.250 0。尼里/拉菲种公牛13 342位点有3种基因型AA、AG、GG,频率分别为0.300 0,0.375 0,0.325 0;13 355位点有3种基因型TT、TG、GG,频率分别为0.575 0,0.325 0,0.100 0;13 406位点产生3种基因型AA、AT、TT,频率分别为0.575 0,0.325 0,0.100 0;17 221位点有3种基因型TT、TC、CC,频率分别为0.600 0,0.325 0,0.075 0。尼里/拉菲种公牛检测群体4个SNP位点均处于HardyWeinberg平衡状态(P>0.05)。 To analyze the genetic polymorphisms of gonadotropin- releasing hormone receptor( GnRHR) gene in Murrah and Nili- Ravi breeding bulls,28 Murral breeding bulls and 40 Nili- Ravi breeding bulls were used for the detection of genetic polymorphisms of GnRHR gene using PCR amplification,sequencing and high- resolution melting( HRM) assay. The results showed that there were same single nucleotide polymorphism( SNP) loci of GnRHR gene between Murrah and Nili- Ravi breeding bulls,including A13 342 G,T13 355 G,A13 406 T in intron1; and T17 221 C in exon 3,which led to the amino acid substitution of Y( tyrosine) for H( histidine). There were the difference genotypes of GnRHR between Murrah and Nili- Ravi breeding bulls. Murrah breeding bulls had two genotypes( AA and AG) at 13 342 locus,and their genotype frequencies were 0. 607 1 and 0. 329 2,respectively; there were two genotypes( TT and TG) at 13 355 locus,and their genotype frequencies were 0. 714 3 and 0. 285 7,respectively; there were two genotypes( AA and AT) at 13 406 locus,and their genotype frequencies were 0. 750 0 and 0. 250 0,respectively; there were two genotypes( TT and TC) at 17 221 locus,and their genotype frequencies were 0. 7500 and 0. 250 0,respectively. Nili- Ravi breeding bulls had three genotypes( AA,AG,and GG) at 13 342 locus,and their genotype frequencies were 0. 300 0,0. 375 0,and 0. 325 0,respectively; there were three genotypes( TT,TG,and GG) at 13 355 locus,and their genotype frequencies were 0. 575 0,0. 325 0,and 0. 100 0,respectively; there were three genotypes( AA,AT,and TT) at 13 406 locus,and their genotype frequencies were 0. 575 0,0. 325 0,and 0. 100 0,respectively; there were three genotypes( TT,TC,and CC) at 17 221 locus,and their genotype frequencies were 0. 600 0,0. 325 0,and 0. 075 0,respectively. Chisquare test showed four SNP loci detected in the group of Nili- Ravi breeding bulls were in Hardy- Weinbery equilibrium( P > 0. 05).
