摘要
目的探讨槲皮素对骨质疏松大鼠骨代谢和股骨生物力学的影响。方法选取SPF级雌性3月龄SD大鼠,60只,随机分成6组,每组各10只,分别为:假手术组(SHAM)、模型组(OVX)、高、中、低剂量槲皮素组(分别称L、M、H组);乙烯雌酚组(E组)。适应性喂养1周后,除SHAM组外,其余各组均进行双侧卵集切除术,术后1周给予药物干预。高、中、低剂量槲皮素组每日按照:200、100、50 mg/(kg·d)剂量,给予槲皮素混悬液灌胃;E组每日按照1.0 mg/(kg·周)剂量给予乙烯雌酚灌胃;SHAM、OVX组给予同等体积生理盐水灌胃。用药后第6、12周分2批各处死半数大鼠,观察并记录各组大鼠血清的骨源性碱性磷酸酶(BALP)、骨钙素(BGP)及雌二醇(E2)的代谢情况。分离右侧股骨进行三点弯曲试验,检测股骨生物力学性能。结果 1)各组血清BGP较OVX组均呈明显增加(P<0.05),并且L、M、H组BGP增加趋势与给予槲皮素剂量呈正相关;与OVX组比较,用药12周较6周时,SHAM、L、M、H组BGP均明显增加(P<0.05),E组无明显变化(P>0.05);2)用药6、12周时,与OVX组内比较,除E组BGP明显增加外(P<0.05),其余各组BALP较OVX组差别无统计学意义(P>0.05);随着用药时间延长,BALP存在减少趋势,但统计学意义不明显;3)而E2的测定结果与BALP相似。只有E组无论是组内比较还是组间比较,较OVX组均明显减少(P<0.05);4)各组大鼠右股骨三点弯曲测试结果差异性不显著。结论槲皮素可以显著影响骨质疏松大鼠的骨代谢,促进成骨,但对骨生物力学性能的影响不显著。
Objective To study the quercetin bone metabolism of osteoporosis rats and the impact of the femur biomechanics. Methods Select SPF female SD rats,3 months only 60,randomly divided into 6 groups,each group of10,only is respectively: control group(SHAM),model group(OVX),high,medium and low dose quercetin group(L,M,H group) respectively. Has female phenol group(group E). Adaptive feed after 1 week,in addition to the SHAM group,the rest of the group were performed bilateral egg collection of resection,postoperative 1 week for drug intervention. High,medium and low dose quercetin group according to the daily: 200,100,50 mg /(kg·d) dose,given quercetin suspension liquid lavage; Group E daily according to 1. 0 mg /(kg · w) dose has ene female phenol lavage;SHAM OVX,given the same volume of saline lavage. 6 and 12 weeks after medication in 2 batch of each executed half rats,observe and record each group the bone of rats serum alkaline phosphatase(BALP),osteocalcin(BGP),estradiol(E2) of metabolism. Separation of right femur in three point bending test,test the femur biomechanics properties. Results Each group serum BGP were significantly increased in the OVX group(P 0. 05),BGP increase trend and L,M,H group and quercetin dose were positively correlated; Compared with OVX group,drug use is 6 weeks,12 weeks BGP SHAM,L,M,H group were significantly increased(P 0. 05),no obvious change in group E(P 0. 05); Use at6 weeks,12 weeks,compared with OVX group,in addition to the BGP E group was obviously increased(P 0. 05),the rest of the group BALP no statistically significant difference in the OVX group(P 0. 05); As the medication time extended,BALP decreasing trend,but no obvious statistical significance; E2 similar to BALP test results. Only group E comparison between whether it is still within the group,were significantly reduced in the OVX group(P 0. 05);Groups of rats right femur three point bending test results no significant differences. Conclusion Quercetin can significantly affect bone metabolism of osteoporosis rats,promote osteogenesis,but no significant effect on the properties of bone biomechanics.
