摘要
以菘蓝(Isatis indigotica)无菌苗的叶片和叶柄为外植体,分别接种到添加不同生长调节剂浓度的MS培养基上,研究不同生长调节剂配比对菘蓝愈伤组织诱导与分化的影响,建立菘蓝组培再生体系。结果表明,叶片是菘蓝愈伤组织形成的最佳外植体,菘蓝愈伤组织诱导的最佳培养基为MS+6-BA 1.0 mg/L+NAA 0.5 mg/L,愈伤组织分化的最佳培养基为MS+6-BA 1.5 mg/L+NAA 1.0 mg/L。以1/2MS培养基添加0.1 mg/L NAA可使再生植株的生根率达到92.3%。
To study the effects of growth regulator on callus induction and differentiation,so that establish tissue culture regeneration system of Isatis indigotica,the leaves and petioles of Isatis indigotica aseptic seedings were cut and inoculated on MS medium supplemented with different growth regulator. The results showed that the leaf is the optimum explant for callus formation of Isatis indigotica and the optimum medium for callus induction and differentiation is MS+6-BA 1.0 mg / L+NAA 0.5mg / L,MS+6-BA 1.5 mg / L+NAA 1.0 mg / L,respectively. When the shoots were cultured on 1 / 2MS medium supplemented with0.1 mg / L NAA, the rooting rate could reach 92.3%.
