有氧运动和茶多酚对2型糖尿病大鼠胰岛素抵抗和血清炎症因子的影响 被引量:4

The Effect of Aerobic Exercise and Tea Polyphenols on Insulin Resistance and Inflammation Factors in Rat with Type 2 Diabetes

摘要 探讨有氧运动和茶多酚对2型糖尿病(T2DM)大鼠胰岛素抵抗和血清炎症因子的影响.采用4周高糖高脂饮食加小剂量注射链脲佐菌素(STZ)(30 mg·kg-1)构建T2DM大鼠模型.实验共5组,分别为正常对照组(C组)、T2DM对照组(DC组)、T2DM运动组(DE组)、T2DM茶多酚组(DT组)、T2DM运动加茶多酚组(DET组).运动组大鼠进行1 h·d-1,每周6 d的无负重游泳训练,茶多酚组以600 mg·kg-1的剂量进行灌胃.8周后取血测空腹血糖、胰岛素及炎症因子TNF-α、IL-6和CRP水平.结果显示:与DC组比较,DE组、DT组和DET组大鼠空腹血糖和胰岛素抵抗指数均显著下降(P<0.05或P<0.01),胰岛素仅DET组下降有显著性(P<0.05);DET组与DE组、DT组比较,空腹血糖、胰岛素和胰岛素抵抗指数均进一步降低,但仅胰岛素抵抗指数的值与DE组有显著差异(P<0.01).与DC组比较,DE组、DT组和DET组大鼠血清TNF-α和CRP水平均显著性降低(P<0.05或P<0.01),而IL-6仅DT组的下降有显著差异(P<0.05);DET组与DE组、DT组比较,仅TNF-α有进一步下降的趋势(P>0.05).胰岛素抵抗指数与血清炎症因子TNF-α和IL-6水平之间呈正相关(P<0.01).表明有氧运动和茶多酚能改善T2DM大鼠的胰岛素抵抗,其机制可能与炎症因子TNF-α和IL-6水平的下降有关,两者联合干预的叠加效果不太明显,但有进一步改善的趋势. This research aimed to explore the effect of aerobic exercise and tea polyphenols on insulin resistance and inflammation factors in rat with type 2 diabetes( T2DM). T2 DM rat model were estabished by injecting a small dose( 30 mg · kg^- 1) of streptozotocin( STZ) into the rats which feeding with high sugar and high fat food for 4 weeks. The experimental rats were randomly divided into 5 group,namely,a normal control group( group C),a T2 DM control group( group DC),a T2 DM exercise group( group DE),a T2 DM TP group( group DT),a T2 DM exercise plus TP group( group DET). The rats in all exercise groups went swimming 6 times per week for 8weeks. The rats in TP adminstration groups were orally administrated TP with the dosage of 600mg·kg^- 1per day for 8 weeks. The fasting blood glucose,insulin and the levels of serum TNF-α、IL-6 and CRP were determined. Compared with group DC,the fasting blood glucos and HOMA-Rdecreased significantly in groups DE,DT and DET( P〈0. 05 or P〈0. 01),and the fasting insulin decreased significantly only in group DET( P〈0. 05). Compared the rats in group DET with the rats in groups DE and DT,the fasting blood glucose,insulin and HOMA-IR decreased further,but HOMA-IR decreased significantly in group DE( P〈0. 01). Compared with group DC,the levels of TNF-α and CRP decreased significantly in groups DE,DT and DET( P〈0. 05 or P〈0. 01),while the level of IL-6 decreased significantly only in group DT. Compared the rats in group DET with the rats in groups DE and DT,the level of TNF-α decreased further( P〉0. 05).HOMA-IR were positively correlated with the levels of serum inflammatory factors,such as TNF-αand IL-6( P〈0. 01). The findings indicated both aerobic exercise and tea polyphenols can improve insulin resistance of T2 DM rats,while may be relate to the decreasing of inflammation factors,including TNF-α and IL-6. The effects of aerobic exercise and tea polyphenols combined intervention are not better than aerobic exercise or tea polyphenols alone,but it has a tendency to improve.
