摘要
为了解天津地区猪繁殖与呼吸综合征病毒(PRRSV)流行毒株的遗传变异情况和流行趋势,用RT-PCR对2014年—2015年在天津地区采集的疑似PRRS病料检测,并扩增阳性病料PRRSV NPS2部分基因片段和进行PRRSV分离鉴定。结果显示,237份疑似PRRSV感染病料中64份为阳性,阳性率为27%,分离获得33株PRRSV;Nsp2序列分析发现,天津地区流行的PRRSV主要为美洲型,其中54份与高致病性猪繁殖与呼吸综合征病毒(HP-PRRSV)高度同源,8份与经典PRRSV高度同源,2份与NADC30-like高度同源。结果表明,2014年—2015年天津地区主要流行HP-PRRSV,并出现了NADC30-like变异毒株,说明PRRSV流行种类的多样化,提示加强PRRSV流行及变异监测十分必要。
This study aimed at analyzing the genetic variation and the development trend of PRRSV in Tian- jin from 2014 to 2015. 237clinical samples collected from 2014 to 2015 in different districts of Tianjin were tested by RT-PCR to amplify partial Nsp2 and isolate virus using positive samples. The result showed that 64 samples were positive, the positive rate was 27%, and acquired 33 PRRSV isolates were field; partial Nsp2 gene and phylogenetic analysis revealed that most of pandemic strains were the North American prototype,and 54 samples were highly homologous with the highly pathogenic PRRSV(HP-PRRSV),8 samples were highly homologous with classic PRRSV, 2 samples were highly homologous with North American pandemic strain NADC30. The study indicated that HP-PRRSV was the popular strain in Tianjin during 2014-2015, the NADC30-like PRRSV emerged make the category of PRRSV variant more diverse,pointing that it is important to pay more attention to monitor prevalent and genetic variation of PRRSV.
