摘要
目的:观察苦瓜总皂苷对2型糖尿病肾病大鼠肾组织中细胞因子信号传导抑制蛋白1(Suppressor of Cytokine Signaling,SOCS1)表达的影响,探讨其对肾脏的保护作用。方法:雄性SD大鼠(48只)随机分为正常对照组、2型糖尿病肾病模型组以及苦瓜总皂苷治疗组5 mg/(kg·d)治疗,左肾切除+1次大剂量链脲佐菌素腹腔注射建立动物模型,模型建立成功后,正常对照组和模型组给予生理盐水灌胃,苦瓜总皂苷治疗组给予苦瓜总皂苷灌胃,分别于实验第4、8周处死大鼠各半,测血糖、24 h尿蛋白定量以及炎性因子;免疫组化、RT-PCR以及Western Blot检测肾脏组织中SOCS1的变化。结果:治疗组尿蛋白和血清肌酐与模型组相比明显减少,血糖明显升高(P<0.05);治疗组与模型组相比,IL-1α、IL-6和TNF-α明显降低,差异有统计学意义(P<0.05);HE染色显示正常组肾脏组织结构整齐,纹理清晰,模型组肾小球基底膜增厚同时伴有一定的系膜细胞增生,苦瓜总皂苷可以明显降低肾脏的损害;免疫组化、RT-PCR和Western Blot可见:与模型组相比治疗后,苦瓜总皂苷治疗组大鼠肾脏组织SOCS1表达明显降低(P<0.05)。结论:苦瓜总皂苷可以通过抑制糖尿病肾病大鼠肾组织SOCS1的表达,进而降低炎症反应,改善肾功能,最终调节2型糖尿病肾病的症状,可能有一定的临床应用价值。
Objective : To investigated the influence of total bitter gourd saponins on type 2 diabetic nephropathy rat suppressor of cytokine signalingl (sacs1)expression, to provided some references for total bitter gourd saponins. Methods : Male SD rats 48 were randomly divided into normal control group, type 2 diabetic nephropathy model group and total bitter gourd saponins ( TBGS ) treatment group ( 5 mg/kg/d ), rat left kidney resection and low-dose streptozotocin intraperitoneal injection built model, after a successful model, control group and model group were given normal saline common 8 weeks/TBGS group was given orally TBGS treated, respectively, after 4. weeks and 8 weeks, half of rats we taken to measure blood sugar, 24 h urinary protein ; the renal tissue of renal pathology was observed with HE staining, RT-PCR and Western Blot to detected the SOCS1 protein and mRNA expression in kidney tissue. Results : Urine protein and serum creatinine of TBGS treated group were decreased, Compared with the same period model group decreased, blood glucose was significantly increased, the difference was significant ( P〈0.05 ); Treatment group compared with model group, IL-1 α , IL-6 and TNF-α were decreased obviously, the difference was statistically significant (P〈0.05); HE stainning can been seen normal control kidneys neat organizational structure, texture clear, model group membrane thickening and mesangial cell proliferation, extracellular matrix significant proliferation, TBGS could significantly improve this feature; hnmunohistochemistry, RT-PCR and Western Blot visible : TBGS group after 4 weeks and 8 weeks treatment, kidney tissue of rats in model group SOCS1 protein and mRNA expression were significantly increased ( P〈0.05 ). Conclusion : TBGS can inhibit renal tissue of diabetic nephropathy rats SOCS1, and protect the kidneys, maybe have some clinical value.
