摘要
目的观察抗纤灵对5/6肾切除小鼠肾组织转化生长因子-β(TGF-β)、结缔组织生长因子(CTGF)表达的影响。方法随机从50只C57小鼠中取40只建立5/6肾切除慢性肾衰肾纤维化模型,术后2周根据肌酐值将40只小鼠分为抗纤灵低、中、高剂量治疗组和模型组,每组10只。另10只小鼠为假手术组。治疗12周后,用western blot法、RT-q PCR法分别测定各组小鼠肾组织TGF-β、CTGF蛋白及mRNA表达水平。结果模型组小鼠肾组织TGF-β、CTGF蛋白及mRNA表达较假手术组升高(P<0.01);与模型组比较,抗纤灵中、高剂量组小鼠肾组织TGF-β及CTGF蛋白、mRNA表达下调(P<0.01),两剂量组间TGF-β、CTGF的蛋白及mRNA表达差异无统计学意义(P>0.05)。结论抗纤灵中、高剂量治疗组均能下调5/6肾切除小鼠肾组织TGF-β、CTGF蛋白及mRNA表达,两剂量治疗组间尚不存在量效相关性。提示抗纤灵可能通过调控TGF-β及其下游因子延缓肾纤维化,保护残肾功能。
Objective To observe the effects of Kangxianling on the expression of transforming growth factor-β ( TGF-β ) and connective tissue growth factor(CTGF) in 5/6 nephrectomy mouse. Methods 40 out of 50 C57 mice were randomly chosen to establish 5/6 nephrectomy mouse model with chronic renal failure and renal fibrosis. Two weeks after the operation, 40 mice were divided into Kangxianling low-dose, middle-dose, high-dose treatment groups and model group according to the value of serum creatinine. The other 10 mice were treated as sham operation group. 12 weeks after the treatment, the expression of TGF-β and CTGF protein in renal tissues of mice of all groups were determined by western blot and RT-qPCR. Results The expression of TGF-β, CTGF protein and mRNA of model group were significantly increased compared with that of the sham operation group (P 〈 0. 01 ). Compared with the model group, the expression of TGF-β, CTGF protein and mRNA were obviously decreased in Kangxianling middle-dose and high-dose groups(P 〈 0. 01 ), but there was no significant difference in the expression of TGF-β, CTGF protein and mRNA between the two groups(P 〉 0. 05 ). Conclusion In Kangxianling middle-dose and high-dose groups, the expression of TGF-β, CTGF protein and mRNA can be lowered in 5/6 nephrectomy mouse, but there is no dose-effect correlation between the two groups. It is suggested that Kangxianling may delay renal fibrosis by regulating TGF-β and its downstream factors and protecting remnant renal function.
