油菜花叶病毒山西地黄分离物的全基因组序列测定与分析 被引量:10

Identification and analysis of complete genomic sequence of Youcai mosaic virus Rehmannia glutinosa isolate in Shanxi

摘要 采用双链RNA(double-stranded RNA,dsRNA)技术和非序列依赖PCR扩增(sequence-independent amplification,SIA)方法对感病地黄进行分子鉴定,并测定油菜花叶病毒(Youcai mosaic virus,YoMV)山西地黄分离物(YoMV-SX)的基因组全序列。序列测定及分析发现侵染地黄的病毒为油菜花叶病毒(Youcai mosaic virus,YoMV)。获得YoMV-SX(Gen Bank登录号JX422022)全长为6 304 nt,5'UTR长度为68 nt,3'UTR长度为236 nt,含有4个开放阅读框(open reading frame,ORF)。全序列核苷酸一致性分析显示Yo M V-SX与Tobamovirus亚组Ⅲ中分离物的一致性为90.7%-96.0%,与同属亚组Ⅰ和Ⅱ的一致性仅为50.2%-63.3%。全序列系统进化分析表明,YoMV-SX与YoMV-Wh形成一个独立分支,亲缘关系最近。这是YoMV侵染地黄的首次报道。 To identify the viruses that caused Rehmannia glutinosa disease, genomic dsRNA was extracted, followed by sequence-independent amplification (SIA) and sequencing. Results showed that there is Youcai mo- saic virus (YoMV) in diseased Rehmannia glutinosa leaves.To further characterize the Shanxi isolate of Youcai mosaic virus (YoMV-SX),the complete sequence of YoMV-SX was cloned and determined.The genomic RNA of YoMV-SX is 6 304 nucleotides in length and presents a typical Tobamovirus genome organization with four ORFs. Sequence alignment of complete nucleotide sequence showed that SX isolate shares the highest identities of 90.7% to 96.0% with that of YoMV isolates belonging to the subgroup II1 of Tobamovirus, and shares low identifies of 50.2% to 63.3% with that of viruses in subgroup I and 11. Phylogenetic analysis based on nucleotide sequences of YoMV-SX and other Tobamovirus isolates showed that YoMV-SX was most closely related to Yo- MV-Wh, and these two isolates formed an independent branch, whereas less relation between YoMV-SX and the isolates within subgroup I and II was observed. Altogether, Rehmannia glutinosa isolate was taxonomically con-sidered as an isolate of YoMV. Our study provides the first report of YoMV in Rehmannia glutinosa.
