胃癌干细胞抗原负载树突细胞与细胞因子诱导的杀伤细胞 被引量:1

摘要 背景:目前临床常用的治疗手段难以彻底消灭肿瘤,其原因主要是由于肿瘤干细胞的存在。目的:探讨胃癌干细胞抗原负载树突细胞结合细胞因子诱导杀伤细胞对胃癌细胞的影响。方法:分离人胃癌细胞系侧群细胞,利用冻融法制备肿瘤抗原,将胃癌干细胞与树突细胞、树突细胞-杀伤细胞、胃癌细胞抗原、胃癌干细胞抗原进行联合培养,采用MTT检测胃癌细胞杀伤率,记录各组树突细胞表面成熟标志CD83表达率。结果与结论:(1)树突细胞组、树突细胞-杀伤细胞组、胃癌细胞抗原组的树突细胞表面成熟标志CD83表达率均显著低于胃癌干细胞抗原组,差异均有显著性意义(P<0.05);(2)树突细胞组、树突细胞-杀伤细胞组、胃癌细胞抗原组的胃癌细胞杀伤率均显著低于胃癌干细胞抗原组,差异均有显著性意义(P<0.05);(3)结果表明,以胃癌干细胞作为抗原刺激树突细胞,并结合细胞因子对杀伤细胞进行诱导,可以促进细胞增殖以及提高胃癌细胞的杀伤能力。 BACKGROUND: As the existence of tumor stem cells, it is difficult to completely eliminate tumors in clinic. OBJECTIVE: To explore the tumor-killing effect of gastric cancer stem cells as antigen to stimulate dendritic cells combined with cytokine-induced killer cells. METHODS: Side population cells from human gastric cancer cell lines were isolated, and tumor antigen was prepared by freeze thawing method. After coculture with dendritic cells, dendritic cells combined with cytokine-induced killer cells, gastric cancer cell antigen, and gastric cancer stem cell antigen, killing rates of gastric cancer cells were detected using MTT assay. Expression rate of CD83, a mature dendritic cell surface marker, was also detected. RESULTS AND CONCLUSION: The CD83 expression level and killing rate of gastric cancer cells were both significantly lower in the gastric cancer stem cell antigen group than the other groups(both P〈 0.05). These results indicate that gastric cancer stem cells as antigen to stimulate dendritic cells combined with cytokine-induced killer cells can promote the proliferation of gastric cancer cells and elevate the ability to killing gastric cancer cells.
