卵丘细胞STAG3表达对卵母细胞及胚胎发育的预测研究

摘要 目的分析卵丘细胞黏连素表达水平与卵母细胞及胚胎发育的关系,探讨卵丘细胞STAG3基因检测用于反应卵母细胞及胚胎发育情况的有效性。方法提取35名患者的IVF-ET周期卵丘细胞,并应用RT-PCR技术定量检测卵丘细胞黏连素STAG3 mRNA的表达水平,分析其与母体因素、卵母细胞发育情况及卵裂率、受精率、优胚形成率、移植周期取消率的关系。结果年龄35岁组的卵丘细胞STAG3表达水平明显低于<35岁组,且差异具有统计学意义(P<0.05);卵丘细胞STAG3表达相对水平>1.00组的患者的Gn刺激天数、Gn总量明显低于卵丘细胞STAG3表达相对水平<1.00组的患者(P<0.05)。STAG3表达上调与卵裂率、受精率、呈现正相关,与移植周期取消率负相关。结论 STAG3基因表达水平与卵母细胞及胚胎发育的存在相关性,对其表达水平的检测具有无创、量化的优势,可以预测卵母细胞及胚胎发育的发育情况。 Objective To analyze the relationship between the expression level of adhesion on cumulus cell and the development of oocytes and embryo, and to explore the effectiveness of mirror oocytes and embryo development with genetic testing for STAG3 on cumulus cell. Methotis The cumulus cell in IVF-ET cycle of 35 patients was extracted, and the expression level of adhesion of STAG3 mRNA on cumulus cell was quantitatively tested by RT-PCR technique,and its relations with maternal factors, oocyte development, fertilization rate, cleavage rate, embryo formation rate, and transplant cycle cancellation rate was analyzed. Results The STAG3 expression level on cumulus cell of patients more than 35-year-old was lower than patients less than 35-year-old, and the difference was statistically significant ( P 〈 0.05 ). The Gn stimulative days and the total Gn amount in patients of the STAG3 relative expression level on cumulus cell great than 1.00 significantly lower than those less than 1 ( P 〈 0. 05 ). The STAG3 high expression was positively correlated with fertilization rate and cleavage rate, while it was negatively related to the transplant cycle cancellation rate. Conclusion The expression levels of STAG3 gene is correlated with the development of oocytes and embryo. The STAG3 gene detection is useful to predict the development of oocytes and embryo.
