苦瓜总皂苷对Ⅱ型糖尿病大鼠心肌细胞凋亡因子Fas和FasL表达的影响 被引量:7

摘要 目的探讨苦瓜总皂苷对Ⅱ型糖尿病大鼠心肌细胞凋亡细胞Fas和FasL表达的影响。方法 60只雄性SD大鼠随机分为正常组、Ⅱ型糖尿病组(模型组)及苦瓜总皂苷治疗组,每组20只。高脂饮食加一次性链脲佐菌素建立Ⅱ型糖尿病大鼠模型,模型建立成功后,治疗组大鼠给予苦瓜总皂苷20mg/kg·d灌胃治疗,其余两组给予生理盐水灌胃参考,分别于治疗4周和8周后处死大鼠各半,测定血糖、乳酸脱氢酶(LDH)、肌酸激酶(CK)以及心肌组织中Fas和FasL的表达特点。结果模型组血糖、LDH以及CK明显高于正常组(P<0.05);治疗组治疗4周后血糖LDH和CK明显降低,治疗8周后进一步降低(P<0.05),免疫组化、Western Blot及RT-PCR可见;模型组Fas和FasL蛋白及mRNA明显高于正常组,苦瓜总皂苷治疗4周后表达明显下降,治疗8周后进一步降低(P<0.05)。结论苦瓜总皂苷可通过抑制Ⅱ型糖尿病大鼠心肌细胞Fas和FasL的表达,进而减轻糖尿病大鼠心肌细胞凋亡,可能对糖尿病早期心脏损害有一定的延缓或治疗作用。 Objective To investigate the effect of total saponins from bitter gourd to Fas and FasL inl heart of type 2 diabetic rat. Methods 60 male SD rats were randomly divided into the normal control group, type 2 diabetes and total saponins from bitter treatment groups. Every group included 20 rats. High-fat diet for one months and streptozotocin established the type 2 diabetic rat model. The treatment group was given total saponins (20mg/kg/d oral) . The remaining two groups were given water fed. 4 weeks and 8 weeks later, the rats were sacrificed. The glucose, lactate dehydrogenase (LDH), creatine kinase (CK) and the expression of Fas and FasL was researched. Results The blood glucose, LDH and CK of model group were significantly higher than that of the control group (P〈0.05). After 4 weeks of treatment, blood glucose LDH and CK of treatment were significantly lower than that of the model group, (P〈0. 05), The Fas and FasL protein and mRNA expression of model group were higher than thaf of the intervention group (P 〈 0.05). Conclusion Saponins can improve the expression of Fas and FasL in type 2 diabetic rat, reduce cardiac myocyte apoptosis, delay early heart damage and produces some therapeutic effect.
