摘要
目的建立高效液相色谱法(HPLC )法测定清火胶囊中芦荟大黄素、大黄素和大黄酚含量的方法.方法:采用 Eclipsc XDB( 4. 6 mm× 250 mm , 5 μm )色谱柱; 流动相为甲醇-0. 1 %磷酸溶液(80:20), 流速为0. 8 0 mL /min, 检测波长:254 nm温30°C .结果:芦荟大黄素、大黄素和大黄酚与其相邻质峰能完全分离.芦荟大黄素、大黄素和大黄酚的线性范围分别0.63-3. 5μg /mL ( r =0.999 9)、0.34-17. 2μg /mL ( r =1.000 0)、0.72-36. 0μg /mL ( r =0.999 8).方法回收率均不于97%.结论:本方法简便、准确、重现性好,能排除其他成分的干扰,可用于该制剂的质量控制的评价.
Objective To establish HPLC method for quantification of aloe-emodin, emodin and chry- sophanol in Qinghuo capsule. Methods: HPLC column was Eclipse XDB (4. 6 mm×250 mm, 5 μm) with methanol-0. 1 % phosphoric acid (80 : 20) as mobile phase. The flow rate was set to 0. 8 mL/min and the wavelength for detection was 254 nm. The column temperature was 30C. Results: Peaks of aloe-emodin, emodin and chrysophanol could be completely isolated from other impurities. The linear range for aloe-emodin, emodin and chrysophanol were 0. 63 to 31. 5μg/mL ( r = 0. 999 9) ,0. 34 to 1 7. 2 μg /mL ( r = 1. 000 0) and 0. 72 to 36. 0 0μg/mL ( r = 0. 999 8), respectively. The recovery rates of all three compounds were not less than 97%. Conclusion: The HPLC method established here is simple, reliable and reproduti- ble,which can be used in the quality control of Qinghuo capsule.
