摘要
〔目的〕改良小鼠视网膜剥离及铺片方法,优化组织学观察及电生理学研究。〔方法〕选用C57BL/6J小鼠在立体显微镜下进行视网膜剥离铺片,然后用免疫荧光和Dil散射标记对其效果进行评估。〔结果〕改良后的方法能够较好地进行视网膜剥离铺片。用免疫荧光染色和Dil散射标记后,包括血管和神经细胞在内的许多组织学结构都能非常形象地观察到。〔结论〕经过改良的视网膜铺片技术能够应用于组织学观察或电生理学研究。
〔Objective〕We tried to improve the methods of retinal detachment and stretched slice in mice.〔Methods〕The C57 BL /6J mice were utilized to detach mouse retina and to prepare retina-stretched slice under dissection microscope, and in the meantime, the effects were evaluated with immunofluorescent labeling and Dil diolistic assay. 〔Results〕The modified methods can be used to detach retina and prepare stretched slice with satisfied results. With immunofluorescent labeling and Dil diolistic assay, the histological structure, including the vessels and neurons, could be visualized well. 〔Conclusion 〕The detached and stretched retina can be applied in the researches of histology and electrophysiology.
