摘要
采用动物蛋白酶对牡蛎蛋白质进行酶解,将得到的牡蛎酶解液通过3k D超滤膜进行分离,探讨超滤过程中操作压力、温度及料液质量浓度对超滤膜通量的影响,确定最佳超滤条件,并测定最佳超滤条件下获得的活性肽抑制卵巢癌SKOV3细胞的增殖能力。结果表明,最佳的超滤工艺条件为:操作压力0.18MPa、温度25℃、料液质量浓度40g/L。在此条件下获得的牡蛎活性肽含量为85.2±0.05%(氮含量为12.7%),其在5mg/L^80mg/L范围内牡蛎活性肽可抑制SKOV3的增殖,24h的半数抑制浓度(IC50)为38.56mg/L。
The ultra-filtration separation process of oyster bioactive peptides and its anti-tumor activity were studied. The hylrolysate derived from oyster enzymatic degradation with animal protease was separated through 3 KD ultra-filtration membrane. The impact of operating pressure, mass concentration of feed solution, ultra-filtration time and temperature were analyzed, and he optimum ultra-filtration conditions were determined. Furthermore, the capacity of bioactive peptides less than 3 KD inhibiting proliferation of ovarian cancer SKOV3 cell was measured. The results showed that the optimum processing conditions of ultra-filration were the pressure with 0.18 MPa, mass concentration of feed solution with 40 g/L and temperature with 25℃. Under the onditions, the content of oyster bioactive peptides was 85.2±0.05% ( 12.7% nitrogen). The oyster bioactive peptides within mg/L-80 mg/L could inhibit the proliferation of SKOV3 with the 50% inhibitory concentration ( IC50 ) 38.56 mg/L.
