摘要
目的:探讨党参多糖对脑缺血缺氧损伤大鼠的保护作用及其机制。方法建立缺血缺氧脑损伤大鼠模型,电镜观察脑组织细胞超微结构病理变化,采用比色法检测脑组织中乳酸脱氢酶(LDH)释放量,ELISA法测定血清中炎症因子白细胞介素-1β(IL-1β)、肿瘤坏死因子(TNF)-α含量。结果相比于模型组,党参多糖干预后显著改善大鼠脑组织细胞完整性,LDH释放量减少、血清IL-1β和TNF-α含量明显降低。结论党参多糖对大鼠脑缺血缺氧损伤具有一定的保护作用,其作用机制可能与抑制炎症反应有关。
Objective To investigate the effects of codonopsis pilosula polysaccharide on the rats with hypoxic-ischemic brain damage and the possible mechanism involved.Methods First,the hypoxic-ischemic brain damage rat model was developed.Pathologic changes in the ultra-structure of the rat brain tissue were closely observed under electron microscopy.The level of lactate dehydrogenase(LDH)released from the brain tissue was detected by colorimetry,and the levels of inflammatory cytokine interleukin-1β(IL-1β)and tumor necrosis factor-α(TNF-α)were detected by ELISA.Results As compared with the model group,codonopsis pilosula polysaccharide intervention could significantly improve the integrity of the rat brain tissue,reduce the LDH level and obviously decrease the levels of IL-1βand TNF-α.Conclusions Codonopsis pilosula polysaccharide has certain protective effects on the rats with hypoxic-ischemic brain damage, and the mechanism involved might be associated with the inhibition of inflammatory response.
