摘要
为研究大蒜素在鱼体内的吸收、消除规律,以7.5 mg/kg(体质量)单剂量口灌给药后,探究大蒜素在松浦镜鲤Cyprinus carpio Songpu体内的药物代谢动力学及药物残留消除规律。试验水温为(20.0±0.5)℃,试验鲤体质量为(1.3±0.2)kg,在给药后0.25、0.50、0.75、1.00、1.50、2.00、4.00、6.00、8.00、12.00、24.00、36.00、48.00、72.00 h采集鲤的血浆,用气相色谱法测定鲤血浆中药物的浓度,并用药动学DAS 3.0软件进行数据分析和处理。结果表明:单次口灌给药后,大蒜素在血浆中的药时关系符合一级吸收二室开放模型;每天一次连续3 d给药后,大蒜素的主要成分二烯丙基二硫化物(DADS)和二烯丙基三硫化物(DATS)均于第15天开始低于10μg/kg。研究表明,在本试验条件下,建议休药期不低于15 d。
The pharmacokinetics and residues of allicin were studied in Songpu mirror carp Cyprinus carpio Songpu exposed to oral administration at a dose of 7. 5 mg/kg( body weight) at water temperature of( 20. 0 ± 0. 5) ℃ by means of gas chromatography( GC). The drug concentrations in plasma were monitored 0. 25,0. 50,0. 75,1. 00,1. 50,2. 00,4. 00,6. 00,8. 00,12. 00,24. 00,36. 00,48. 00 h,and 72. 00 h after drug administration,and analyzed by DAS 3. 0 software. The results showed that the concentrations of allicin in muscle versus time were well described by two-department open model with first-order absorption after a single oral administration. The residues in Songpu mirror carp by oral administration for 3 d once a day was found to be less than the maximal residue limit( MRL)( 10 μg/kg) at the day 15 for DADS and DATS. The findings indicate that the withdrawal time should be15 d after administration under these conditions.
